敲减CCDC132表达对宫颈癌HeLa细胞生长和凋亡的影响

摘要 目的分析敲减CCDC132表达对宫颈癌HeLa细胞生长和凋亡的影响。方法以CCDC132-shRNA(shCCDC132组)和阴性对照Ctrl-shRNA (shCtrl组)慢病毒感染人宫颈癌HeLa细胞后,采用荧光显微镜观察HeLa细胞感染效率,应用实时荧光定量PCR检测HeLa细胞中CCDC132 mRNA表达水平,应用Western blot检测HeLa细胞中CCDC132蛋白表达水平,应用Celigo细胞计数法检测HeLa细胞生长情况,应用流式细胞仪检测HeLa细胞凋亡情况,应用Caspase-3活性检测试剂盒检测HeLa细胞Caspase-3活性。结果 shCtrl组宫颈癌HeLa细胞中CCDC132 mRNA和蛋白表达结果分别为(1.00±0.11)和(0.21±0.03),shCCDC132组宫颈癌HeLa细胞中CCDC132 mRNA和蛋白表达结果分别为(0.42±0.03)和(0.05±0.01)。与shCtrl组比较,shCCDC132组CCDC132 mRNA和蛋白表达水平均显著降低(t=15.261,P <0.001;t=15.179,P <0.001)。与shCtrl组比较,shCCDC132组细胞生长明显受到抑制(均P <0.05)。shCtrl组宫颈癌HeLa细胞凋亡率和Caspase-3相对活性分别为(6.36±1.15)%和(1.00±0.05),shCCDC132组宫颈癌HeLa细胞凋亡率和Caspase-3相对活性分别为(17.54±2.62)%和(3.86±0.12),与shCtrl组比较,shCCDC132组细胞凋亡率和Caspase-3活性均显著升高(t=11.722,P<0.001;t=66.000,P<0.001)。结论敲减CCDC132表达可抑制宫颈癌HeLa细胞生长,诱导细胞凋亡。
