芹菜叶斑病病原菌的分离鉴定、生物学特性及其生防菌筛选 被引量:20

摘要 为了明确甘肃省定西市芹菜叶斑病的病原,开展了病原菌分离、形态学特征和分子生物学鉴定、生物学特性研究,并初步筛选了该病原菌的生防菌。确定病原菌Q1为细极链格孢(Alternaria tenuissima),该菌菌丝体生长最适碳源和氮源分别为玉米粉和硝酸钾,最适pH为7.5,最适温度为25℃;该菌孢子萌发最适温度为25℃、最适pH为7;光照有利于菌丝体生长,但对孢子萌发无影响;菌丝体和孢子的致死温度均为55℃处理10 min。采用对峙法测定了蜡样芽胞杆菌(4-2-1)、贝莱斯芽孢杆菌(F3A)、甲基营养型芽孢杆菌(9-2-1)、解淀粉芽孢杆菌(SL1)及荧光假单胞菌(YG)5种生防细菌菌株对芹菜叶斑病菌Q1的拮抗效果,结果表明这5株生防菌对Q1菌株均具有拮抗作用,其中解淀粉芽孢杆菌(Bacillus amyloliquefaciens)的抑菌率最高,达到69.80%。首次明确了定西市芹菜叶斑病是细极链格孢(A.tenuissima)引起的,解淀粉芽孢杆菌(SL1)具有生防潜力,研究结果为深入开展芹菜链格孢叶斑类病害的生物防治提供了理论依据和技术支撑。 In order to identify the pathogens which cause leaf spot of celery in Dingxi,Gansu province,the isolation,morphological and molecular biological identification and biological characteristics of the pathogens were carried out,and the bio-control of the pathogens was preliminarily studied.The isolated and identified pathogen Q1 was Alternaria tenuissima.The optimal carbon and nitrogen sources for the growth of the mycelia were maize meal and potassium nitrate,respectively.The optimal temperature and pH was 25℃and 7.5,respectively.Light could promote the growth of mycelia,but had no effect on spore germination.The lethal temperature of mycelia and spores was 55℃for 10 min.The confrontation method was used to determine the antagonistic effect of biocontrol bacteria strains against celery leaf spot fungus Q1,including Bacillus cereus(4-2-1),B.velezensis(F3A),B.methylotrophicus(9-2-1),B.amyloliquefaciens(SL1)and Pseudomonas fluorescens(YG).The results showed that all the 5 bio-control strains had antagonistic effects on Q1,among them the bacteriostatic rate of SL1 was highest,reaching 69.80%.This study confirmed that celery leaf spot disease in Dingxi was caused by A.tenuissima for the first time,and B.amylolyticus(SL1)had the potential to develop biological pesticide.The results provide theoretical basis and technical support for further study on control of celery leaf spot disease.
