黑曲霉孢子性状与糖化力的相关性分析 被引量:1

Correlation analysis of spore trait and saccharification power of Aspergillus niger

摘要 该试验从麸曲中分离纯化鉴定出10株黑曲霉,比较了不同黑曲霉种曲的颜色及产孢量,用水解圈和3,5-二硝基水杨酸(DNS)法定性定量比较其糖化力,进而分析了黑曲霉孢子性状与糖化力的相关性。结果表明,菌株AW1、N3、D11、D2种曲产孢量较多,分别为9.55×10^(9)个/g、9.35×10^(9)个/g、8.97×10^(9)个/g、8.8×10^(9)个/g;菌株AY1和AY2种曲产孢量较少,为1.95×10^(8)个/g、4.13×10^(8)个/g;扫描电镜(SEM)比较菌株AW1、N3、AY1和AY2的分生孢子头,前两者分生孢子头大且小梗紧密,后两者分生孢子头小且小梗疏松;菌株AY1和N3的D/d值及糖化力最高,分别为2.36、1.29,3158.2 U、2991.9 U;糖化力定性和定量结果比较吻合,但与黑曲霉孢子的数量和颜色没有明显的相关性。 Ten strains of Aspergillus niger were isolated and purified from Fuqu,the color and spore yield of different A.niger Zhongqu were compared,the saccharifying power was compared qualitatively and quantitatively by hydrolytic circle and 3,5-dinitrosalicylic acid(DNS)method,and then the correlation between saccharification power and spore trait of A.niger was analyzed.The results showed that the spore yield of strains AW1,N3,D11 and D2 was high,which were 9.55×10^(9) spore/g,9.35×10^(9) spore/g,8.97×10^(9) spore/g and 8.8×10^(9) spore/g,respectively.The spore yield of strains AY1 and AY2 were low,which were 4.13×10^(8) spore/g and 1.95×10^(8) spore/g,respectively.The conidia head of strains AW1,N3,AY1 and AY2 were compared by scanning electron microscope(SEM).The conidia head of strains AW1 and N3 was large and tight,and the conidia head of strains AY1 and AY2 was small and loose.The D/d value and saccharifying power of strains AY1 and N3 were the highest,which were 2.36,1.29,and 3158.2 U,2991.9 U,respectively.The qualitative and quantitative results of saccharification power were similar,but there was no significant correlation with the number and color of A.niger spores.
