外源NO对高温胁迫下茄子幼苗生长和抗氧化系统的影响 被引量:7

Effect of Exogenous NO on the Plant Growth and Antioxidant System of Eggplant Seedlings under High Temperature Stress

摘要 为探讨外源一氧化氮供体硝普钠(SNP)缓解高温胁迫对茄子幼苗伤害的可行性,以茄子(Solanum melongena L.)品种‘特旺达’为试验材料,在人工气候箱内,研究了0.05-5 mmol/L的SNP对高温胁迫(43±1)℃/(38±1)℃(昼温/夜温)下茄子幼苗生长和活性氧代谢系统的影响。结果显示:(1)高温显著抑制了茄子幼苗的生长,且显著降低了叶绿素含量;0.1 mmol/L SNP处理下,茄子幼苗的生长促进效果最明显,叶绿素含量升高幅度最大。(2)用0.1 mmol/L SNP处理显著降低了高温胁迫下的茄子幼苗丙二醛(MDA)含量、超氧阴离子(O2·-)产生速率和过氧化氢(H2O2)含量;同时茄子幼苗中的渗透性物质(脯氨酸,可溶性蛋白,可溶性糖)含量显著增加。(3)茄子幼苗在高温胁迫下的超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、过氧化物酶(POD)和抗坏血酸过氧化物酶(APX)、脱氢抗坏血酸还原酶(DHAR)、单脱氢抗坏血酸还原酶(MDHAR)、谷胱甘肽还原酶(GR)等抗氧化酶活性及抗氧化物质抗坏血酸(AsA)和还原型谷胱甘肽(GSH)含量经过0.1 mmol/L SNP处理后,均显著增加,As A/DHA、GSH/GSSG比值降低。添加NO清除剂血红蛋白部分或完全逆转了上述作用。本研究表明,在高温胁迫下的茄子幼苗经0.1 mmol/L SNP处理,其抗氧化酶活性和渗透调节物质含量显著提高,对细胞膜系统的稳定性有维持作用,因此缓解了高温胁迫下茄子幼苗的氧化损伤,增强了茄子的耐热性。 In order to investigate the feasibility of exogenous nitric oxide donor sodium nitroprusside(SNP)in alleviating the damage to eggplant seedlings under high temperature stress,the eggplant variety’Tewangda’was used as the experimental material.In the artificial climate chamber,the effects of 0.05~5 mmol/L SNP on the growth and reactive oxygen metabolism of eggplant seedlings under high temperature stress(43±1)℃/(38±1)℃(day/night)were studied.The results showed that:(1)High temperature significantly inhibited the growth of eggplant seedlings and significantly reduced the content of chlorophyll.Under the treatment of 0.1 mmol/L SNP,the growth promotion effect of eggplant seedlings was the most obvious,and the increase of chlorophyll content was the largest.(2)The content of malondialdehyde(MDA),superoxide anion(O2·-)production rate and hydrogen peroxide(H2O2)in eggplant seedlings under high temperature stress were significantly decreased after 0.1 mmol/L SNP treatment.Meanwhile,the content of permeable substances(proline,soluble protein,soluble sugar)increased significantly in eggplant seedlings.(3)The superoxide dismutase(SOD),catalase(CAT)and peroxidase(POD)and ascorbic acid peroxidase(APX),dehydrogenation ascorbic acid reductase(DHAR),single dehydrogenation ascorbic acid reductase(MDHAR),glutathione reductase(GR)activities of antioxidant enzymes and antioxidants such as ascorbic acid(AsA)and glutathione(GSH)contents of eggplant seedlings under high temperature stress,after dealing with the 0.1 mmol/L SNP,were significantly increased.However,the As A/DHA,GSH/GSSG ratio were reduced.The addition of NO scavenger hemoglobin partially or completely reversed the above effects.This study showed that the activity of antioxidant enzymes and the content of osmotic regulating substances in eggplant seedlings treated with 0.1 mmol/L SNP under high temperature stress were significantly increased,and the stability of cell membrane system was maintained.Therefore,the oxidative damage of eggplant seedlings under high temperature stress was alleviated and the heat resistance of eggplant was enhanced.
