摘要
目的白菜在运输和储存过程中容易腐败变质,许多不良商家纷纷添加价格低廉的甲醛用来防腐,鉴于目前食品风险监测均是大批量样品测定,建立一种快速、简便的测定白菜中甲醛含量的酚试剂衍生分光光度检测方法非常必要。方法样品粉碎均匀后,以水作溶剂,40℃超声提取30 min,利用甲醛与酚试剂发生衍生反应,在酸性条件下被高铁离子氧化成蓝绿色化合物,其颜色深浅与吸光度大小成一定比例,630 nm处比色测定。结果甲醛浓度在0μg~4.5μg内线性良好,r=0.999 6,检出限为0.4 mg/kg。本底阴性样品中分别添加高、中、低3个浓度进行加标测定,回收率为95.6%~102.6%,RSD<4.0%。结论本方法灵敏度高、重现性好,方法快速简便、结果准确可靠,特别适合基层实验室的应用。
Objective Cabbage is prone to spoilage during transportation and storage,many unscrupulous merchants added low-cost formaldehyde for preservative purposes.At present,food risk monitoring is measured by large quantities of samples.Therefore,a fast and simple method using the phenol reagent-derived spectrophotometric for determining the formaldehyde content in cabbage is very necessary.Methods After the sample is evenly crushed,it is extracted with water as a solvent and ultrasonically extracted at 40 ℃ for 30 min.Formaldehyde is derived with a phenol reagent.Under acidic conditions,it is oxidized by ferric ion to a blue-green compound.The color depth is proportional to the absorbance.Colorimetric measurement is used at 630 nm.Results The formaldehyde concentration was linear within 0 μg-4.5 μg(r=0.999 6),and the detection limit was 0.4 mg/kg.Three concentrations of high,medium,and low were added to the negative samples for determination.The recovery was within 95.6%-102.6%,and the RSD was less than 4.0%.Conclusion In addition to simplicity,the method is accurate reproductive and reliable,which is especially suitable for the application of primary laboratories.
