植物生长调节剂对鞑靼忍冬扦插生根及酶活性变化的影响 被引量:17

摘要 【目的】鞑靼忍冬抗逆性强,为西北干旱半干旱区重要的园林观赏植物之一,应用前景广阔。以萘乙酸(NAA)、吲哚丁酸(IBA)和吲哚乙酸(IAA)3种生长调节剂不同浓度、不同处理时间处理鞑靼忍冬扦插穗条进行扦插繁殖试验,以期筛选出促进鞑靼忍冬插穗生根的最佳处理组合,同时明晰扦插生根过程中过氧化物酶(POD)、多酚氧化酶(PPO)和超氧化物歧化酶(SOD)活性的变化对生根指标的影响,为西北干旱半干旱区鞑靼忍冬扦插快繁提供理论依据。【方法】以鞑靼忍冬当年生嫩枝插条为材料,利用正交试验探讨生长调节剂的种类、浓度和处理插穗的时间等因素对鞑靼忍冬插穗生根的影响及不定根发生过程中POD、PPO和SOD活性的变化及其与插穗生根之间的关系。【结果】鞑靼忍冬插穗生根类型为皮部生根型以及少量的愈伤组织生根型;根系发育进程可分为不定根原基诱导期、不定根形成期、不定根伸长期、不定根快速生长和增粗期;正交试验各处理的生根指标除最小根长差异不显著外,其余指标均显著优于对照组(P<0.05);T4(50 mg/L IBA处理1 h)和T6(200 mg/L IBA处理30 min)综合生根效果较好;IBA处理组对POD、PPO和SOD活性均比IAA处理组和NAA处理组效果明显,且IBA处理组中,T4对POD、PPO和SOD活性的效果最佳。【结论】综合分析得出:IBA处理组中T4(50 mg/L IBA处理1 h)为最佳处理组。 【Objective】Lonicera tatarica,with strong resistance to stress,is one of the important garden ornamental plants in the arid and semi-arid regions of northwest China.The plant has a broad application prospect.The cutting propagation test of Lonicera tatarica cuttings was carried out with three growth regulators of NAA,IBA and IAA at different concentrations and different treatment time,in order to screen out the optimal treatment combination to promote the rooting of Lonicera tatarica cuttings,and the effects of changes in peroxidase(POD),polyphenol oxidase(PPO)and superoxide dismutase(SOD)enzyme activities on rooting indicators,so as to provide a theoretical basis for the rapid propagation of Lonicera tatarica cuttings in the arid and semi-arid areas of northwest China.【Method】Using the cuttings of Lonicera tatarica as the material,the orthogonal test was used to explore the influence of the type and concentration of growth regulators and the time of processing the cuttings on the Lonicera tatarica cuttings rooting and adventitious roots occur in the process of peroxidase(POD),polyphenol oxidase(PPO)and superoxide dismutase(SOD)activity of change and the relationship between the cuttings to take root.【Result】The rooting type of Lonicera tatarica cuttings is cortical rooting.The rooting process can be divided into adventitious root primordium induction period,adventitious root formation period,adventitious root extension period,adventitious root rapid growth and thicken period.In the orthogonal experiment,the rooting indexes of each treatment are not significantly different except the minimum root length,the other indexes are significantly better than the control group(P<0.05)and the T4 group(treated with 50 mg/L IBA for 1 h)and T6(treated with 200 mg/L IBA for 30 min)had better comprehensive rooting effect.The effect of IBA treatment group in peroxidase(POD),polyphenol oxidase(PPO)and superoxide dismutase(SOD)activity was significantly better than that of IAA and NAA treatment group.In addition,the activities of peroxidase(POD),polyphenol oxidase(PPO)and superoxide dismutase(SOD)in T4 group were more obvious than those in other groups.【Conclusion】The comprehensive analysis shows that T4(50 mg/L IBA treatment for 1 h)treatment group in IBA treatment group is the best treatment group.
