摘要
目的探究解郁汤对脑卒中后抑郁(PSD)模型大鼠行为学及对5-HT、NE含量表达的影响。方法SD大鼠60只,随机分成6组,采用经典的线栓大脑中动脉阻塞法造模,加用不可预见温和应激,建立大鼠PSD模型。造模成功3 d后,予以药物干预,实验周期为21 d。末次给药后,测量大鼠体质量,进行蔗糖水实验、敞箱实验,并采用酶联免疫分析(ELISA)法检测大鼠脑组织5-羟色胺(5-HT)、去甲肾上腺素(NE)。结果与模型组相比,西药组、解郁汤高、中、低剂量组体质量均明显增加(P<0.05);西药组体质量与解郁汤高剂量组相比,差异无统计学意义(P> 0.05);说明解郁汤高剂量组在改善PSD大鼠体质量方面更具优势。西药组、解郁汤高、中、低剂量组糖水消耗量较模型组明显增加(P<0.05),解郁汤高剂量组与西药组比较,糖水消耗量差异无统计学意义(P> 0.05),说明解郁汤高剂量组与盐酸氟西汀在改善糖水消耗方面具有等效性。敞箱实验中,西药组、解郁汤高、中剂量组水平活动、垂直活动得分均较模型组明显提高(P<0.05);与西药组比较,解郁汤高、中剂量组大鼠水平活动得分、垂体活动得分差异无统计学意义(P> 0.05),说明解郁汤高、中剂量组与盐酸氟西汀在改善PSD大鼠自发行为方面具有等效性。脑中5-HT、NE含量,西药组、解郁汤高、中、低剂量组均高于模型组(P<0.05),说明解郁汤可影响脑卒中后脑5-HT、NE的表达,其中以高剂量组效果最为明显。结论解郁汤可明显改善脑卒中后抑郁大鼠的行为,提高卒中后抑郁模型大鼠脑5-HT、NE含量,且以高剂量组效果最为显著。
Objective To explore the effect of Jieyu decoction(解郁汤) on behavior and the expression of 5-HT and NE inPSD rats.Methods60 SD rats were randomLy divided into 6 groups. The model of PSD was established by the classical methodof middle cerebral artery occlusion and unpredictable mild stress. After 3 days of successful modeling, drug intervention wasgiven. Blank group: given the same volume of distilled water by gavage;model group:given the same volume of distilled waterby gavage;western medicine group: given fluoxetine hydrochloride by gavage. Jieyu decoction high dose group(37. 8 g/kg,141. 6%, 2 mL/100 g). Jieyu decoction medium dose group(18. 9 g/kg,70. 8%, 2 mL/100 g). Jieyu decoction low dosegroup(9. 45 g/kg, 35. 4%, 2 mL/100 g). Rats in each group were intragastrically administered once a day for 21 days. Afterthe experiment, body weight was measured, open box test and sucrose water test were carried out,5-HT,NE in rat brain tissue were detected by ELISA.ResultsAfter the experiment, the body weight of Western medicine group, Jieyu decoction high-dose group, middle dose group and low-dose group increased significantly(P< 0. 05);there was no significant differencebetween western medicine group and Jieyu Decoction high-dose group(P> 0. 05);it showed that Jieyu Decoction highdose group had more advantages in improving the body weight of PSD rats. After the experiment, the consumption of sugar water in western medicine group, Jieyu Decoction high-dose group, middle dose group and low-dose group increased signifi-cantly(P< 0. 05). Compared with western medicine group, the consumption of sugar water in Jieyu Decoction high-dosegroup had no significant difference(P> 0. 05);the consumption of sugar water in Jieyu decoction middle dose group and low-dose group were lower than that in western medicine group(P< 0. 05),indicating that Jieyu decoction high-dose groupand Prozac have equivalence in improving sugar water consumption. The scores of horizontal activity and vertical activity inwestern medicine group, Jieyu decoction high-dose group and middle dose group were increased(P< 0. 05);compared withwestern medicine group, there was no significant difference in the scores of horizontal activity and pituitary activity in JieyuDecoction high-dose group and middle dose group(P> 0. 05);the scores of horizontal activity and pituitary activity in JieyuDecoction low-dose group were lower than those in western medicine group(P< 0. 05), indicating that Jieyu Decoction high-dose group and middle dose group had no significant difference with western medicine group(P> 0. 05) Baiyou solution isequivalent in improving the spontaneous behavior of PSD rats. Compared with western medicine group, 5-HT and ne of JieyuDecoction high-dose group and low-dose group were higher than those of model group(P< 0. 05);compared with westernmedicine group, 5-HT and ne of Jieyu decoction high-dose group had no significant difference(P> 0. 05);5-HT and neof Jieyu Decoction medium dose group and low-dose group were lower than those of Western medicine group(P< 0. 05),which indicated that Jieyu decoction could affect the expression of 5-HT and NE, and the effect of high-dose group was themost obvious.ConclusionJieyu decoction can significantly improve the behavior of post-stroke depression rats and increasethe levels of 5-HT and NE in the model rats, especially in the high-dose group.
