SASH1在小细胞肺癌中的表达及临床意义

Expression and clinical significance of SASH1 in small cell lung cancer

摘要 目的探讨候选抑癌基因SASH1在小细胞肺癌(SCLC)组织中的表达及其与临床病理特征关系。方法收集临沂市肿瘤医院2012年2月至2018年2月手术切除并经病理学诊断确诊的SCLC癌组织及癌旁组织石蜡标本各30例,应用免疫组织化学法检测SCLC癌组织及癌旁组织中SASH1蛋白的表达,通过SPSS 20.0统计学软件对数据进行χ2检验,分析SASH1蛋白在SCLC中的表达及其与临床病理特征之间的关系。结果SASH1蛋白在癌组织阳性表达率为43.33%,显著低于癌旁组织的86.67%,差异有统计学意义(P<0.001)。在免疫组化检测的30例SCLC中,男性组SASH1蛋白的阳性表达率为45.83%(11/24),女性组为33.33%(2/6),差异无统计学意义(P>0.05);<60岁组SASH1蛋白的阳性表达率为50.00%(6/12),≥60岁组为38.89%(7/18),差异无统计学意义(P>0.05);SCLC伴淋巴结转移组阳性表达率为30.00%(6/20),低于不伴淋巴结转移组的70.00%(7/10),差异有统计学意义(P<0.05);肿瘤长径≥3 cm的SASH1阳性表达率为43.48%(10/23),<3 cm的阳性表达率为42.86%(3/7),差异无统计学意义(P>0.05)。结论SASH1基因在SCLC中表达下调,该基因可能是SCLC的抑癌基因,并且与是否出现淋巴结转移相关,与性别、年龄、肿瘤大小无关。 Objective To investigate the expression of candidate tumor suppressor gene SASH1 in small cell lung cancer(SCLC)and its relationship with clinicopathological features.Methods A total of 30 paraffin specimens of SCLC tissues and 30 adjacent tissues were collected from patients who underwent surgical resection and confirmed as SCLC by pathological diagnosis in Linyi Cancer Hospital from February 2012 to February 2018.The expressions of SASH1 protein in SCLC carcinoma and adjacent tissues were detected by immunohistochemical method.The data were tested byχ2 test with SPSS 20.0 statistical software,and the expressions of SASH1 protein in SCLC tissues and its relationship with clinicopathological features were analyzed.Results The positive expression rate of SASH1 protein in cancer tissues was 43.33%,which was significantly lower than that in paracancerous tissues(86.67%),and the difference was statistically significant(P<0.001).In 30 cases of SCLC detected by immunohistochemistry,the positive expression rate of SASH1 protein was 45.83%(11/24)in the male group and 33.33%(2/6)in the female group,and there was no significant difference between the two groups(P>0.05).The positive expression rate of SASH1 protein was 50.00%(6/12)in the<60 years old group and 38.89%(7/18)in the≥60 years old group,and there was no significant difference between the two groups(P>0.05).The positive expression rate of SCLC in the group with lymph node metastasis was 30.00%(6/20),which was lower thanthat in the group without lymph node metastasis(70.00%,7/10)(P<0.05).The positive expression rate of SASH1 in the group with tumor diameter≥3 cm was 43.48%(10/23),and the positive expression rate in the group with tumor diameter<3 cm was 42.86%(3/7).There was no significant difference between the two groups(P>0.05).Conclusion The expression of SASH1 gene is down-regulated in SCLC,which may be a tumor suppressor gene of SCLC and is related to lymph node metastasis,but not to gender,age and tumor size.
