摘要
该研究建立了气相色谱-质谱联用仪(GC-MS)同时测定植物蛋白饮料中β-谷甾醇、豆甾醇、菜籽甾醇、菜油甾醇、豆甾烷醇5种植物甾醇及胆固醇含量的方法。在优化升温程序、柱流速等仪器条件基础上,对比确定了样品取样量、皂化碱液浓度及最佳的衍生化条件。样品经皂化、提取、浓缩并衍生化后用正已烷定容,经GC-MS选择离子(SIM)扫描模式分析,5α-胆甾烷内标法定量。经方法学验证,各甾醇组分方法检出限为0.032~0.058 mg/kg,定量限为0.104~0.190 mg/kg,标准曲线线性关系良好(R2≥0.9999),在0.304~21.3 mg/kg质量浓度范围内,加标回收率为90.6%~101.2%,相对标准偏差(RSD)≤5.3%。
A method for the simultaneous determination of 5 phytosterols and cholesterol(β-sitosterol,stigmastol,rapesesterol,rapesesterol,stigmastanol)in vegetable protein drinks was developed by GC-MS.On the basis of optimized temperature programming,column flow rate and other instrument conditions,the sample amount,saponification alkali concentration and derivatization conditions were improved.After saponification,extraction,concentration and derivatization,the samples were measured by n-hexane,analyzed by GC-MS in selective ion monitor(SIM)and quantified by 5α-cholestane as internal standard.The results showed that the detection limit of sterols was 0.032-0.058 mg/kg,and the limit of quantification was 0.104-0.190 mg/kg,the standard curves had good linearity(R2≥0.9999).Within the range of 0.304-21.3 mg/kg,the standard recovery was 90.6%-101.2%,and the relative standard deviation(RSD)was less than 5.3%.
