恒山黄芪种子中抗菌肽提取条件的优化和抑菌率测定 被引量:6

Optimization of Extraction Conditions of Antimicrobial Peptides from Astragalus Seeds and Determination of Antibacterial Rate

摘要 目的:对黄芪种子抗菌肽提取条件进行优化,提高提取率并测定抑菌率。方法:以黄芪种子为材料,使用双指数响应面分析法对其提取条件进行优化,使用96孔板法测定抑菌率。结果:黄芪种子抗菌肽的最佳提取条件:浸提液为EDTA缓冲液,料液比为1:5 g/mL,浸提时间为16.5 h,硫酸铵质量百分比浓度为82%。此工艺下,黄芪种子抗菌肽的抑菌率可达80.3%,比优化前(75.6%)提高了4.7%;提取率可达79.6%,较优化前(67.2%)提高了12.4%。结论:采用双指数响应面分析法对黄芪种子抗菌肽的提取条件的优化有效,并且抑菌率也有很大提升。 Objective:To optimize the extraction conditions of antimicrobial peptides from Astragalus seeds,improve the extraction rate and determine the antibacterial rate.Methods:Using Astragalus seeds as material,the extraction conditions were optimized by double exponential response surface analysis,and the inhibition rate was determined by 96-well plate culture.Results:The optimal extraction conditions of antimicrobial peptides from Astragalus seeds were as follows:the extract was EDTA buffer,the material-liquid ratio was 1:5 g/mL,the extraction time was 16.5 h,and the ammonium sulfate saturation was 82%.The antibacterial rate of Astragalus seed antimicrobial peptides can reach 80.3%,which is 4.7% higher than that before optimization (75.6%).The extraction rate is 79.6%,which is an increase of 12.4% compared to before preoptimization (67.2%).Conclusion:The double exponential response surface analysis is effective to optimize the extraction conditions of antimicrobial peptides from Astragalus seeds,and the antibacterial rate is also greatly improved.
