摘要
目的对贵州省2006-2016年分离的炭疽芽胞杆菌进行单核苷酸重复序列分型分析(SNR),了解菌株SNR型别特征,为炭疽疫情监测、调查与溯源提供技术手段和科学依据。方法提取贵州省2006-2016年间不同地区的炭疽芽胞杆菌菌株基因组DNA,应用SNR各位点引物进行PCR扩增,将扩增产物进行毛细管电泳分析获得各位点序列长度。运用Bionumerics 7.6软件将扩增长度片段信息进行聚类分析获得各菌株SNR型别并构建最小间距图。结果来自贵州省2006-2016年间的71株炭疽芽胞杆菌被分为37个SNR型;聚类分析显示,71株菌株被分为A和B两群,其中A群共有27个分支,又可分为A1和A2两个亚群;B群共有10个分支,可分为B1和B2两个亚群。最小间距图显示,71株菌株被分别为7个SNR克隆群(complex)和5个单一的SNR型(singleton)。结论来自贵州省2006-2016年间的71株炭疽芽胞杆菌共分为37个SNR型,SNR型别具有多样性;SNR分子分型方法对炭疽芽胞杆菌分型具有高分辨率的特点,SNR型别分布与菌株的时间和地域分布具有一定相关性。
To provide a technical means and scientific basis for the investigation,surveillance,and source tracking of anthrax,single-nucleotide repeat analysis(SNR)was applied to analyze Bacillus anthracis isolated in Guizhou Province from 2006 to 2016.The genomic DNA of Bacillus anthracis isolates from samples from different regions of Guizhou Province was extracted and amplified with PCR primers targeting SNR loci.Then the sequence length of the amplification product of each SNR locus was obtained through capillary electrophoresis.The SNR type was obtained,and the clustering tree and minimum spanning tree based on the length of each SNR locus of the B.anthracis were constructed using Bionumerics 7.6 software.SNR genotyping showed that 71 isolates of Bacillus anthracis were divided into 37 SNR genotypes.Cluster analysis indicated that 71 strains were clustered into groups A and B.Group A,with a total of 27 clades,could be further divided into two subgroups,A1 and A2.Group B,with a total of ten clades,could be divided into subgroups B1 and B2.The minimum tree showed that 71 strains were distributed in seven SNR clone complexes and five singletons.In conclusion,37 SNR genotypes were identified among the 71 strains of Bacillus anthracis isolated in Guizhou Province from 2006 to 2016,with a diversity of SNR types.The SNR molecular typing method has high resolution for Bacillus anthracis typing.SNR type distribution is correlated with regional and temporal aggregation.
