摘要
目的:建立和评价膝骨关节炎(KOA)肾阳虚大鼠模型。方法:24只大鼠随机分成正常组、KOA组、KOA去势组、KOA腺嘌呤组,每组6只。Hulth法建立KOA模型,去势法与腺嘌呤灌胃建立肾阳虚模型。通过Mankin’s和OARSI评分评价KOA模型;通过肾阳虚症状体征评价表及甲状腺轴、性腺轴、肾上腺轴激素水平等评价肾阳虚模型。结果:KOA组、KOA去势组、KOA腺嘌呤组均符合KOA病理表现,OARSI分级分别为2级、3级和4级。与其余3组比较,KOA腺嘌呤组肾阳虚症状体征积分最高(P<0.01,P<0.05),体质量增长最少(P<0.05),肛温最低(P<0.05),T3、T4、FSH、LH、T水平最低(P<0.05),E2水平最高(P<0.05);各组间CORT水平差异无统计学意义。结论:相较于Hulth+去势法,Hulth+腺嘌呤灌胃是构建KOA肾阳虚病证结合模型的更适宜方法。
Objective:To establish and evaluate the rat model of knee osteoarthritis(KOA)with kidney yang deficiency.Methods:Twenty-four rats were randomly divided into normal group,KOA group,KOA castration group and KOA adenine group,with 6 rats in each group.The KOA model was established by Hulth method,and the kidney yang deficiency model was established by castration method and adenine gavage.The KOA model was evaluated by Mankin’s and OARSI scores;the kidney yang deficiency model was assessed by a form of symptoms and signs of kidney yang deficiency,thyroid axis hormone levels,gonadal axis hormone levels,adrenal axis hormone levels,etc..Results:The KOA group,KOA castration group,and KOA adenine group were all consistent with the pathological manifestations of KOA,with the OARSI classification standard of grade 2,grade 3 and grade 4,respectively.Compared with the other three groups,KOA adenine group had the highest score of symptoms and signs of kidney yang deficiency(P<0.01,P<0.05),the least weight gain(P<0.05),the lowest anal temperature(P<0.05),the lowest levels of T3,T4,FSH,LH and T,and the highest level of E2(P<0.05);there was no significant difference on the level of CORT among groups.Conclusion:Compared with Hulth plus castration,Hulth plus adenine gavage is a more suitable method for the establishment of KOA kidney yang deficiency model.
