摘要
目的:制备三氧化二砷纳米粒并进行细胞学实验,分析体外药效及其对耐药细胞的逆转倍数。方法:运用CCK-8法考察纳米粒的细胞毒性以及纳米粒对乳腺癌细胞MCF-7/ADR多药耐药的逆转;采用Annexin V-FITC/PI双染法,用流式细胞仪检测三氧化二砷纳米粒诱导细胞凋亡的作用;采用PI单染流式细胞术检测As_(2)O_(3)-NPs对MCF-7/ADR周期的阻滞。结果:细胞学实验结果表明,各组药物均能够抑制耐药细胞的正常生长,凋亡率呈浓度依赖性,纳米粒组的凋亡率高于游离药物组;游离药物组和纳米粒组均阻滞细胞生长于G2/M期,As_(2)O_(3)-mPEG-PLA-NPs阻滞效果最好。As_(2)O_(3)、As_(2)O_(3)-PLA-NPs、As_(2)O_(3)-mPEG-PLA-NPs作为逆转剂时,ADM对MCF-7/ADR的逆转倍数分别为1.32、1.8和2.11。结论:As_(2)O_(3)-PLA-NPs、As_(2)O_(3)-mPEG-PLA-NPs均对乳腺癌耐药细胞MCF-7/ADR有促凋亡作用,对G2/M期细胞生长阻滞明显,对MCF-7/ADR的逆转作用强于As_(2)O_(3)。
Objective:To analyze the efficacy of As_(2)O_(3) nanoparticles(As_(2)O_(3)-NPs)in vitro and its reversal multiples for drug resistant cells based on cytological experiments.Methods:CCK-8 method was used to study the toxicity of nanoparticles on cells and the reversal of MCF-7/ADR.Annexin V-FITC/PI double staining and flow cytometry were used to detect As_(2)O_(3)-NPs inducing cell apoptosis.PI staining flow cytometry was used to detect the cycle block of MCF-7/ADR by As_(2)O_(3)-NPs.Results:Cytology results showed that the drug in each group was able to inhibit the normal growth of resistant cells,apoptosis rate showed a concentration-dependent trend,and the apoptosis rate was higher in As_(2)O_(3)-NPs group than that in the free drug group.Both the free drug group and the As_(2)O_(3)-NPs group arrested cells growth in G2/M phase,As_(2)O_(3)-mPEG-PLA-NPs showed the best arrested effect.As_(2)O_(3),As_(2)O_(3)-PLA-NPs,As_(2)O_(3)-mPEG-PLA-NPs,as reversal agents,the reversal multiples of ADM to MCF-7/ADR were 1.32,1.8 and 2.11.Conclusion:Both As_(2)O_(3)-PLA-NPs and As_(2)O_(3)-mPEG-PLA-NPs can promote apoptosis of MCF-7/ADR,which is breast cancer cell with multi-drug resistance,the G2/M phase of cell growth is arrested obviously,and their reverse effect on MCF-7/ADR is stronger than As_(2)O_(3).
