茶多酚-羧甲基壳聚糖水凝胶促进大鼠创面修复研究 被引量:4

Effect of epigallocatechin gallate-carboxymethyl chitosan hydrogel on wound healing in rats

摘要 目的制备一款新型的茶多酚(EGCG)-羧甲基壳聚糖(CMCS)水凝胶,并研究其对大鼠创面的保护和修复作用。方法 45只SPF级健康雄性SD大鼠随机分入随机分入空白组、CMCS水凝胶组、EGCG-CMCS水凝胶组,每组各15只。建立大鼠皮肤全缺损模型,建模成功后,每天给予CMCS水凝胶组和EGCG-CMCS水凝胶组大鼠创面CMCS水凝胶和EGCG-CMCS水凝胶,连续3 d。大鼠皮肤组织分别在术后7、10、15 d取材。结果 EGCG-CMCS水凝胶组创伤恢复百分比高于其他两组(P<0.05),且EGCG-CMCS水凝胶组创面结痂完全脱落,出现新生肉芽组织。EGCG-CMCS水凝胶组可见大量新生肉芽组织和新生毛细血管,表面细胞大量增殖,已经出现较为致密的胶原纤维,炎症细胞基本消失;且EGCG-CMCS水凝胶组的上表皮化水平和胶原蛋白纤维致密水平显著好于其余两组,胶原纤维含量也显著高于其余两组(P<0.05)。结论 EGCG-CMCS水凝胶具有良好的促进血管生成和表皮再生作用,可促进大鼠创面的胶原蛋白沉积和肉芽组织形成,加速创面愈合。 Objective To prepare a new type of epigallocatechin gallate(EGCG)-carboxymethyl chitosan(CMCS)hydrogel and investigate its protective and repair effect on wound in rats.Methods Totally 45 healthy SD male rats were randomly divided into the blank group,CMCS hydrogel group and EGCG-CMCS hydrogel group,with 15 rats in each group.The wound model of full-thickness defect in rats was established.After successful modeling,the wounds of rats in CMCS hydrogel group and EGCG-CMCS hydrogel group were treated with CMCS hydrogel and EGCG-CMCS,for 3 consecutive days,and the skin samples were taken at 7,10 and 15 days after operation.Results The wound healing rate in the EGCG-CMCS hydrogel group was significantly higher than those in the other two groups(P<0.05),and the scab in the EGCG-CMCS hydrogel group had completely fallen off and new granulation tissue appeared.Meanwhile,a large number of new granulation tissue and new capillaries were observed in EGCG-CMCS hydrogel group,and a large number of surface cells proliferated,and more dense collagen fibers appeared.Inflammatory cells basically disappeared,and the level of epidermization and collagen fiber density in EGCG-CMCS hydrogel group was significantly better than those in the other two groups.Conclusion EGCG-CMCS hydrogel has a better effect on promoting angiogenesis and epidermal regeneration,promoting collagen deposition,granulation tissue formation and wound healing in rats.
