摘要
目的从代谢组学角度探讨肝宝胶囊治疗非酒精性脂肪性肝病(NAFLD)的作用机制。方法采用高脂饮食诱导NAFLD大鼠模型,SD大鼠随机分为空白组、模型组、阳性对照组和肝宝胶囊高、中、低剂量组,连续给药4周。ELISA法对大鼠血清ALT(丙氨酸氨基转移酶)、AST(天冬氨酸氨基转移酶)、TC(总胆固醇)、TG(甘油三酯)、LDL⁃C(低密度脂蛋白胆固醇)、HDL⁃C(高密度脂蛋白胆固醇)生化指标进行测定,HE染色观察肝组织病理变化,利用UPLC⁃Q⁃TOF/MS技术并结合PCA(主成分分析)、PLS⁃DA(最小二乘法判别分析)和OPLS⁃DA(正交最小二乘法判别分析)等多元统计分析方法,筛选潜在差异代谢物及其代谢通路分析。结果肝宝胶囊可显著降低NAFLD大鼠血清ALT、AST、TC、TG和LDL⁃C水平,显著增加HDL⁃C水平,使NAFLD大鼠共26个差异代谢物水平发生不同程度回调,主要涉及代谢通路为甘油磷脂代谢、鞘脂代谢与花生四烯酸代谢。结论肝宝胶囊可能通过调节内源性物质代谢紊乱来发挥治疗NAFLD的作用。
AIM To explore the mechanism of Ganbao Capsules in treating non⁃alcoholic fatty liver(NAFLD)by metabolomics approach.METHODS The NAFLD rat model was induced by high⁃fat diet.SD rats were randomly divided into normal group,model group,positive control group,and Ganbao Capsules groups(at high,medium,and low doses)for consecutive administration over four weeks.The rats had their serum levels of ALT,AST,TC,TG,LDL⁃C,HDL⁃C detected by ELISA,and their hepatic pathological changes observed by HE staining.UPLC⁃Q⁃TOF/MS based metabolic profiling method,combined with multivariate statistical analysis,such as PCA,PLS⁃DA and OPLS⁃DA,was conducted to search for potential differential metabolites and to explore related metabolic pathways after treatment of Ganbao Capsules.RESULTS Ganbao Capsules significantly decreased the levels of ALT,AST,TC,TG,and LDL⁃C;increased HDL⁃C level;and had different degrees of callbacks on twenty⁃six differential serum metabolites of NAFLD rats.The metabolic pathways mainly involved with metabolisms of glycerophospholipid,sphingolipid and arachidonic acid.CONCLUSION Ganbao Capsules may play an important role in the treatment of NAFLD by regulating the metabolism disorders of endogenous substances.
