TLR4在抵抗生殖道感染过程中对小鼠卵泡发育的影响 被引量:2

摘要 目的探讨固有免疫调节受体TLR4在抵抗生殖道感染过程中对卵泡发育的影响。方法雌性BALB/c小鼠随机分为LPS组、PBS组和未处理组,每组12只。组织学方法计数各组卵泡发育的变化,real time qPCR技术检测TLR成员在LPS组卵巢中的表达模式。体外培养BALB/c小鼠的卵巢,随机分为LPS刺激组和对照组。利用real time qPCR技术检测卵巢中相关信号分子IL-1β、IL-6、IL-8的表达,利用ELISA检测培养液中E2水平。结果与未处理组相比,LPS组卵巢的原始卵泡数目降低,闭锁卵泡数目升高,差异有统计学意义(P<0.05)。LPS组中,与其他TLR成员相比TLR4在小鼠卵巢中呈现较高水平的表达,差异有统计学意义(P<0.05)。离体实验显示,与对照组相比,LPS刺激组TLR4的下游炎症因子IL-1β、IL-8和IL-6的表达升高,但E2水平降低,差异有统计学意义(P<0.05)。结论LPS使TLR4诱导炎症反应,同时还影响了卵泡发育。这对于发现感染导致的卵泡发育不良的新调控机制具有重要意义。 Objective To investigate the influence of the innate immune regulation receptor TLR4 on oocyte development during reproductive tract infection resistance.Methods BALB/c female mice were randomly divided into LPS group,PBS group and untreated group,with 12 in each group.Histological approach was used to determine the follicle count in each group.In LPS group,the expression patterns of TLR members were detected in ovary by real time qPCR.The ovarian tissue of BALB/c mice was cultured in vivo and then divided into LPS group and control group.Expression of signaling molecules(IL-1β,IL-6 and IL-8)in ovary were quantified by real time qPCR.E2 level in culture media was measured using ELISA.Results Compared with untreated group,the primordial follicles were significantly decreased,while the atretic follicles were significantly increased in LPS group(all P<0.05).Compared with the other TLRs,the expression level of TLR4 in ovary was enhanced significantly in LPS group(P<0.05).In vivo,compared with control group,the expression levels of TLR4 down-stream cytokines(IL-1β,IL-8 and IL-6)were increased significantly,while E2 level in media was decreased significantly in LPS group(all P<0.05).Conclusion TLR4 is not only able to induce the immune response,but also can affect oocyte development in ovary during LPS infection.This study provides a significant insight into the new regulatory mechanisms of follicular dysplasia after infection.
