摘要
目的研究手足口病发病与细胞因子水平异常变化的关系,研究EV71病毒诱导细胞天然免疫的发生机制。方法将Jurkat细胞接种到24孔板中,设EV71病毒感染组和对照组,并用Realtime-PCR法检测感染不同时间段的细胞上清液中IL1-β、IL6、TNF-α、IFN-γ的mRNA表达含量。用乙酰肉豆蔻佛波酯(phorbol-12-myristate-13-acetate,PMA)预作用Jurkat细胞不同时间后,接种EV71病毒,检测细胞上清液中病毒载量变化。结果EV71病毒感染Jurkat细胞后,IL1-β的mRNA在6 h后达峰,IL-6的mRNA在24 h后达峰,TNF-α和IFN-γ的mRNA分别在48 h和72 h达峰;PMA预作用Jurkat细胞后,IL1-β、IL6、TNF-α、IFN-γ的mRNA表达均上调,病毒载量下降。结论随着EV71病毒作用时间的延长,IL1-β、IL6、TNF-α、IFN-γ的mRNA表达均达峰值,Jurkat细胞内EV71病毒载量呈递减趋势。EV71可引起IL1-β、IL6、TNF-α、IFN-γ细胞因子反应。
Objective To find relationship between the onset of hand,foot,and mouth disease and abnormal changes of cytokine levels,so as to clarify the mechanism of enterovirus 71(EV71)-induced natural cellular immunity.Methods Jurkat cells were cultured in 24-well plates and the EV71 infection group and control group were set up.The mRNA expression levels of IL1-β,IL6,TNF-αand IFN-γin supernatants collected at different time after infection were detected by realtime-PCR.EV71 was inoculated into Jurkat cells pre-treated with phorbol-12-myristate-13-acetate(PMA)for different time to analyze the changes of viral loads in supernatant of cell culture.Results After Jurkat cells infected by EV71,the mRNA of IL1-βreached peak level at 6h,the mRNA of IL-6 reached peak level after 24h while the mRNAs of TNF-αand IFN-γreached peak levels after 48h and 72h,respectively.For PMA-pre-treated Jurkat cells,the mRNA expressions of IL1-β,IL6,TNF-αand IFN-γwere all up-regulated with decreased viral loads.Conclusions With the prolongation of infection by EV71,all mRNA expressions of IL1-β,IL6,TNF-αand IFN-γreached peak levels while the viral loads in Jurkat cells decreased.EV71 can induce cytokine reactions,including IL1-β,IL6,TNF-αand IFN-γ.
