高效液相色谱法测定不同杀青工艺咖啡叶茶的主要功能成分

摘要 为充分利用咖啡叶中富含的活性物质,本研究筛选了滚筒、微波和蒸汽3种杀青方式制作咖啡叶茶,在对其进行感官审评的基础上,通过高效液相色谱法(HPLC)检测咖啡碱、葫芦巴碱、儿茶素、芦丁、绿原酸和芒果苷的含量。同时也开展其自由基清除试验。结果表明,滚筒和微波杀青工艺的感官审评得分更高。3种杀青方式咖啡叶中的儿茶素、绿原酸和芒果苷含量差异显著,而生物碱类物质(咖啡碱、葫芦巴碱)含量差别小。滚筒杀青咖啡叶清除DPPH型和ABTS型自由基的能力最强。不同杀青工艺对咖啡叶的功能成分含量影响较大。滚筒杀青咖啡叶的清除自由基能力最强。本研究可为咖啡叶茶的提质加工和咖啡树资源的综合利用提供科技支撑。 Coffee leaves are rich in active substances,which are worthy of being fully utilized.Coffee leaves were processed with three different fixation methods of roller,microwave and steamin this study.Sensory evaluation,detecting the contents of caffeine,trigonelline,catechin,rutin,chlorogenic acid and mangiferin by high performance liquid chromatography(HPLC),and the free radical scavenging tests were carried out to comprehensively evaluate activity of coffee leaf tea.The results showed that the sensory evaluation scores of the rollerfixed and microwave-fixed are higher.The contents of catechin,chlorogenic acid,and mangiferin in coffee leaves with different fixation methods were significantly different,while the contents of alkaloids(Caffeine,trigonelline)was similar.The roller-fixed coffee leaves tea had the strongest scavenging ability on both DPPH and ABTS free radicals.Different curing processes have a greater impact on the contents of functional components of coffee leaves.Roller-fixedcoffee leaves have the strongest free radical scavenging ability.This research can provide scientific and technological support for the quality improvement of coffee leaf tea and the comprehensive utilization of coffee tree resources.
