摘要
为研究糜子麸皮中多酚提取条件及抗氧化活性,利用超声波-微波协同萃取技术,以多酚提取量为指标,在单因素实验基础上,选液料比、乙醇体积分数、提取温度、超声波功率进行响应面实验;并测定糜子麸皮多酚抗氧化活性。结果表明,糜子麸皮中多酚最佳提取工艺条件为:液料比50∶1 mL/g,乙醇体积分数60%,提取温度75℃,超声波功率1000 W,微波功率为200 W,提取时间10 min。在此条件下,糜子麸皮中多酚提取量为8.92 mg/g。抗氧化实验结果显示,该多酚对于DPPH自由基、羟自由基、超氧自由基的清除率和还原力的IC 50值分别为0.006、0.142、12.048、4.022 mg/mL,并且糜子麸皮多酚与上述抗氧化指标间均呈显著正相关(P<0.05),表明糜子麸皮中多酚具有较强的抗氧化和自由基清除能力。
In order to study the extraction conditions and antioxidant activity of polyphenols from millet bran,ultrasonic microwave synergistic extraction technology was used.Based on single factor experiment,extraction yield was used as an index,response surface experiments were carried out with liquid-material ratio,ethanol volume fraction,extraction temperature and ultrasonic power.Meanwhile,the antioxidant activity from millet bran were evaluated.The results indicated that the optimum extraction conditions from millet bran were as follows:the liquid-material ratio was 50∶1 mL/g,the volume fraction of ethanol was 60%,the extraction temperature was 75℃,the ultrasonic power was 1000 W,the microwave power was 200 W and the extraction time was 10 minutes.Under these conditions,polyphenols extraction yield from millet bran was 8.92 mg/g.The results of antioxidant experiments indicated that the IC 50 values of the polyphenols for DPPH free radical scavenging rate,hydroxyl radical scavenging rate,superoxide radical scavenging rate and reducing power were 0.006,0.142,12.048 mg/mL and 4.022 mg/mL,respectively.And the polyphenols indicated significant positive correlation with the antioxidant activity indexes mentioned above(P<0.05).The results indicated that the polyphenols from millet bran have strong antioxidant and free radical scavenging ability.
