摘要
探究苦参止痢颗粒治疗仔猪腹泻的作用机理。通过BATMAN数据库检索苦参止痢颗粒中化学成分与作用靶点,利用STRING 11.0、DAVID数据库和Cytoscape 3.7.2软件从苦参止痢颗粒中筛选出丝裂原活化蛋白激酶1(MAPK1)、TP53、ESR1、PPP2CB、PRKCA、PRKCD、PGR和AR等关键靶点;GO富集发现1条细胞组成条目,2条分子功能条目以及5条生物过程条目;6条KEGG通路,包括心肌细胞中肾上腺素能信号、血管平滑肌收缩、钙信号通路、精氨酸生物合成、唾液分泌和cGMP-PKG信号通路。说明苦参止痢颗粒可通过壬酸、月桂酸、殷金醇棕榈酸酯、22,23-二氢豆甾醇、白桦脂醇以及没食子酸等成分调控基因靶点,并通过信号转导、神经调节、酶活性调节以及内分泌调节等生物过程来调控仔猪腹泻。
The purpose of this study was to investigate the mechanism of sophora flavescens check dysentery granule in treating piglet diarrhea.Searching the chemical constituents and action targets in sophora flavescens check dysentery granule by BATMAN database.Key targets such as MAPK1,TP53,ESR1,PPP2CB,PRKCA,PRKCD,PGR,and AR were selected from sophora flavescens check dysentery granule through STRING 11.0,DAVID database,and Cytoscape 3.7.2 software.GO enrichment found 1 cell composition entry,2 molecular function entries and 5 biological process entries.There are 6 KEGG pathways,including adrenergic signaling in cardiomyocytes,vascular smooth muscle contraction,calcium signaling pathway,arginine biosynthesis,salivary secretion and cGMP-PKG signaling pathway.It was concluded that sophora flavescens check dysentery granule could regulate gene targets through nonanoic acid,lauric acid,20-Hexadecanoylingenol,22,23-dihydrostigmasterol,betulin and gallic acid.Biological processes including signal transduction neurotransmitter,neural regulation,enzyme activity regulation and endocrine regulation to regulate piglet diarrhea.
