摘要
文章以α-葡萄糖苷酶为目标酶,先制备Fe3 O4@PDA@Au磁性纳米粒子,通过Fe3 O4@PDA@Au磁性纳米粒子与α-葡萄糖苷酶间的作用力固定酶分子,建立了磁性酶反应器—高效液相色谱技术在线筛选中草药酶抑制剂的方法。对Fe3 O4@PDA@Au磁性纳米粒子上固定化酶的活性、酶促反应的pH值、反应时间和温度等影响因素进行考察,测定了α-葡萄糖苷酶的米氏常数Km,并对固化酶的结构进行表征。最后,对固化酶在8味中草药粗提物中酶抑制活性成分的筛选进行初步研究。
Study on the preparation of Fe3 O4@PDA@Au magnetic nanoparticles,establishing a screening model of immobilized enzyme for alpha glycosidase inhibitors and building a new method of screening enzyme inhibitors of Chinese herbal medicine by the mean of magnetic immobilized enzyme reactor-HPLC online detection.The main influencing factors are investigated in experiments,which can affect the treatment effects such as activity of immobilized enzymes,reaction temperature,reaction time and pH value.Determine michaelis constant of α-glucosidase(Km),and characterize the structure of curing enzyme by TGA and IR.At last,the model was used to screen the α-glucosidase inhibitors active ingredients from 8 kinds of crude extract of Chinese herbal medicine.
