摘要
为探讨犬间充质干细胞对脊髓损伤治疗的效果,将40只大鼠随机分为空白对照组、假手术组、脂肪间充质干细胞治疗组(SCI+ADMSCs)、骨髓间充质干细胞治疗组(SCI+BMMSCs)和模型对照组(SCI+PBS),其中SCI+ADMSCs组、SCI+BMMSCs组及SCI+PBS组使用改良Allens打击法制备大鼠脊髓损伤模型,造模后SCI+ADMSCs组及SCI+BMMSCs组于1,2,3,7,14 d以1×10^6 mL/kg细胞数量局部多点注射犬间充质干细胞,SCI+PBS组注射等量PBS溶液,空白对照组与假手术组不做注射处理。使用BBB评分法评价大鼠运动功能恢复情况,ELISA及免疫组化法分别检测血浆及脊髓组织炎性因子含量,TUNEL法检测脊髓组织细胞凋亡率,免疫荧光法检测髓鞘及GFAP含量等方法评价犬间充质干细胞对脊髓损伤的治疗作用。结果造模后7,14,21,28 d SCI+ADMSCs组与SCI+BMMSCs组大鼠BBB评分均较SCI+PBS组有极显著性上升(P<0.01);SCI+ADMSCs组与SCI+BMMSCs组大鼠血浆及组织中tnf-α含量较SCI+PBS组显著下降(P<0.05);SCI+PBS组细胞凋亡率及脊髓瘢痕组织含量显著高于其余4组(P<0.05),而脊髓髓鞘含量显著低于其余4组(P<0.05)。结果表明,犬脂肪及骨髓来源的间充质干细胞均可促进大鼠脊髓损伤的修复,其修复作用可能通过降低炎性因子、降低细胞凋亡率、减少瘢痕组织形成以及增加或维持脊髓组织中髓鞘含量等途径实现。
To investigate the effect of canine mesenchymal stem cells on the treatment of spinal cord injury(SCI),40 rats were randomly divided into blank control group,sham operation group,adipose-derived mesenchymal stem cell treatment group(SCI+ADMSCs),bone marrow mesenchymal stem cell treatment group(SCI+BMMSCs)and model control group(SCI+PBS).The SCI+ADMSCs group,SCI+BMMSCs group and SCI+BMMSCs group were used to prepare rat spinal cord injury model by modified Allens method.At 1,2,3,7 and 14 d after modeling,SCI+ADMSCs group and SCI+BMMSCs group were injected the number of canine mesenchymal stem cells(1×10^6 mL/kg).The SCI+PBS group was injected with the same amount of PBS solution.The blank control group and the sham operation group were not injected.The BBB score was used to evaluate the recovery of motor function in rats.The levels of inflammatory factors in plasma and spinal cord were detected by ELISA and immunohistochemistry.The apoptosis of spinal cord tissue was detected by TUNEL.The content of myelin and GFAP was detected by immunofluorescence.All those methods were used to evaluate the repairing effect of canine mesenchymal stem cells on spinal cord injury.The result of BBB scores of SCI+ADMSCs group and SCI+BMMSCs group were significantly higher than those of SCI+PBS group at 7,14,21,28 dafter model establishment(P<0.01).The level of tnf-αin plasma and tissues of SCI+ADMSCs group and SCI+BMMSCs group were significantly lower than those in SCI+PBS group(P<0.05).In the apoptosis rate among groups,SCI+PBS group was significantly higher than that of other groups(P<0.05).The content of GFAP in SCI+PBS group was significantly higher than that in the other 4 groups(P<0.05).The myelin sheath content in the SCI+PBS group was significantly lower than the other 4 groups(P<0.05).This experiment proves that canine adipose-derived mesenchymal stem cells and bone marrow mesenchymal stem cells can promote the repair of spinal cord injury,and its effect is to reduce the expression of inflammatory factors,the rate of apoptosis,the formation of scar tissue and increase or maintain the myelin content in spinal cord tissue.
