高孔隙率海藻酸钠-聚乙烯醇复合海绵的制备及性能 被引量:8

Preparation and Properties of High Porosity Sodium Alginate/Polyvinyl Alcohol Composite Sponges

摘要 以聚乙烯醇(PVA)和海藻酸钠(SA)为原料,诺氟沙星(NFX)为药物模型,采用溶液共混和冷冻干燥成型相结合的方式制备了载药型SA-PVA海绵敷料。采用傅里叶变换红外光谱(FT-IR)和扫描电子显微镜(SEM)等手段对其物化结构进行了表征。研究了产物的吸水性和保水性等物理性能,并对其进行了体外生物相容性及小鼠体内实验评价。结果显示:复合海绵内部呈现三维网络结构,孔隙率最高可达95.1%,具有适宜的吸水性和保水性。另外,以新鲜兔血为实验用血研究了海绵的溶血情况,SA-PVA复合海绵的溶血率均小于2%,无溶血性,敷料对NIH 3T3细胞没有毒性,细胞相对增殖率均大于90%;小鼠体内实验证明,制备的载药型SA-PVA海绵敷料能够有效促进伤口愈合。SA-PVA复合海绵有望作为治疗难愈合创口的敷料使用。 The drug-loaded SA/PVA sponge dressings were prepared by solution blending and freeze-dried molding with polyvinyl alcohol(PVA)and sodium alginate(SA),and norfloxacin(NFX)was used as a drug model.The physicochemical structures were characterized by Fourier transform infrared spectroscopy(FT-IR)and scanning electron microscopy(SEM).The physical properties including the water absorption and water retention of the products were studied.The in vitro biocompatibility and in vivo experiment of the wound dressings were evaluated.The composite sponges exhibited three-dimensional network structure with the porosity of up to 95.1%,and had suitable water absorption and water retention.In addition,the fresh rabbit blood was used as experimental blood to study the hemolysis of sponges,and the results showed the hemolysis of the SA/PVA composite sponges was less than 2%,and the prepared composite sponges had no hemolysis.The relative proliferation rate of the wound dressings was more than 90%,and the SA/PVA composite sponges were non-cytotoxic.From in vivo results,the prepared drug-loaded SA/PVA sponge dressings could effectively promote wound healing.The SA/PVA composite sponges are expected to be used as dressings for the treatment of refractory wounds.
