QuEChERS/超高效液相色谱-串联质谱法同时测定香蕉中8种植物生长调节剂残留 被引量:7

Simultaneous Determination of 8 Plant Growth Regulator Residues in Banana by Using Ultra High Performance Liquid Chromatography-Tandem Mass Spectrometry in Combination with QuEChERS Methodology

摘要 为了测定植物生长调节剂在香蕉中的残留,本研究建立了QuEChERS结合超高效液相色谱-串联质谱法同时测定香蕉中8种植物生长调节剂残留的分析方法。以1%乙酸-乙腈(V/V)为提取溶剂,样品前处理采用QuEChERS方法。以Agilent InfinityLab Poroshell 120 EC-C18柱为分离色谱柱,甲醇和5 mmol/L乙酸铵-0.1%甲酸缓冲溶液为流动相,0.25 mL/min的流速梯度洗脱,10 min内可实现8种目标待测物分离。8种目标待测物经超高效液相色谱-串联质谱在选择反应监测模式下测定,基质匹配标准溶液外标法定量。结果表明,在5.0~100.0μg/kg范围内线性良好,相关系数(r2)≥0.999;检出限(LODs)范围为0.03~0.6μg/kg,定量限范围为0.10~2.0μg/kg。在10、20、100μg/kg 3个添加水平范围内,平均回收率在73.5%~107.2%之间,相对标准偏差(RSD)小于11%。本方法简便、快速、便捷、灵敏、准确,适用于香蕉中8种植物生长调节剂残留的测定。本方法的建立可为植物生长调节剂在其他果蔬中的残留检测提供参考。 In order to determine the residue of plant growth regulators in banana,a method was developed for the simultaneous determination of 8 plant growth regulators residues(PGRs)in banana by using QuEChERS(quick,easy,cheap,effective,rugged and safe)purification coupled with ultra high performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS).Using acetonitrile containing 1%acetic acid(V/V)as extraction solvent,the samples was prepared by the modified QuEChERS method.The chromatographic separation of 8 target compounds was accomplished in an Agilent InfinityLab Poroshell 120 EC-C18 column using methanol and 5 mmol/L ammonium acetate-0.1%formic acid buffer solution as mobile phase by gradient elution at a flow rate of 0.25 mL/min in 10 min.The 8 target compounds were analyzed by using UPLC-MS/MS in selective reaction monitoring(SRM)mode and then were quantified by the matrix matched standard calibration curve method.The method evaluation was done by matrix-matched calibration with linearity ranging from 5 to 100μg/kg with a correlation coefficient more than 0.999.The detection and quantification limit ranged from 0.03 to 0.6μg/kg and 0.10 to 2.0μg/kg,respectively.The average recoveries at three spiked concentration levels of 10,20,100μg/kg for all target compounds in the samples were in the range of 73.5%to 107.2%,with relative standard deviations(RSD)not more than 11%.The method is simple,quick,convenient,sensitive,and can be applied to analyze 8 plant growth regulators in banana.This method will provide a reference for the residue detection of plant growth regulators in other fruits and vegetables.
