摘要
目的:探讨自拟中药防疫香囊预防瘟疫的潜在靶点基因和可能的作用机制.方法:首先,查阅古今医籍文献,追溯中医药香囊辟瘟除秽的发展.然后,借助中药系统药理学分析平台(TCMSP)检索防疫香囊主要挥发性成分相应的靶点基因,通过GeneCards等数据库筛选疾病的靶点基因,应用Cytoscape 3.7.2构建药物挥发性成分-靶点网络图,通过STRING数据库构建靶点基因相互作用网络及富集分析.结果:获得防疫香囊挥发性成分-疾病互作靶点基因共19个;关键靶点基因有FOS、TNF、TP53、PTGS2、IL-10、CDKN1A、RHOA、ADRB2、OPRM1、SLC6A4等,主要富集于细胞信号转导、免疫、IL-4和IL-10等信号通路.结论:防疫香囊可能通过多种活性化合物,作用于多靶点,促进有利于抗病毒的细胞信号转导,提高机体免疫功能,发挥预防瘟疫的作用.
Objective:To explore the potential target genes and possible mechanisms of herbal sachets in preventing epidemic disease.Methods:Firstly,the development of herbal sachet in preventing epidemic disease was excavated from ancient and modern medical literature.Then,traditional Chinese medicine systems pharmacology(TCMSP)platform was used to search the target genes of the volatile components of the Chinese medicine herbal sachet,and the target genes of disease were screened through GeneCards databases.The volatile drug components-target network map was constructed by using Cytoscape 3.7.2,and interaction network of target gene and enrichment analysis was constructed by STRING database.Results:A total of 19 volatile component-disease interaction target genes were obtained for epidemic prevention sachets.The key target genes include FOS,TNF,TP53,PTGS2,IL-10,CDKN1 A,RHOA.,ADRB2,OPRM1,SLC6 A4 and so on,which were mainly enriched in signaling pathways of cell signal transduction,immunity,IL-4 and IL-10.Conclusion:Herbal sachets may act on multiple targets through multiple volatile components,promote anti-viral cell signal transduction,improve the body’s immune function,and thus to play a role in preventing epidemic disease.
