摘要
以子二代中华鲟(Acipenser sinensis)为实验对象,研究不同亚硝酸盐浓度(0.01、0.20、0.40、0.80、1.60 mg·L-1)慢性胁迫(60 d)对鱼体生长、抗氧化及免疫指标的影响。结果显示,高浓度组子二代中华鲟发生死亡现象,死亡率为33.33%,其他各组存活率均为100%;试验鱼的增重率和特定生长率随亚硝酸盐浓度的增加而逐渐降低(P<0.05),而脏体比、肝体比和肥满度无显著变化(P>0.05)。在抗氧化指标中,血清和肝脏超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、总抗氧化能力(T-AOC)活性及谷胱甘肽(GSH)含量随亚硝酸盐浓度的升高而下降,而丙二醛(MDA)含量随亚硝酸盐浓度的升高而上升。免疫指标中,随着亚硝酸盐浓度的升高,血清、肝脏溶菌酶(LZM)活性和免疫球蛋白M(IgM)含量均显著下降(P<0.05)。结果表明,慢性亚硝酸盐胁迫显著抑制了子二代中华鲟的生长,降低了鱼体的抗氧化能力和免疫能力,这对了解中华鲟亚硝酸盐胁迫反应以及其健康养殖具有重要的借鉴意义。
Chinese sturgeon(Acipenser sinensis)is a large-scale upstream spawning fish,which belongs to the national first-class protected animals.However,in the process of artificial breeding of Acipenser sinensis,the dissolution of high-protein residual baits,the discharge of high-nitrogen excretions and the impediment of nitrification can lead to the accumulation of nitrite in water.Nitrite can not only cause a series of physiological reactions such as spasm and hyperventilation,but also severely reduce the oxygen carrying capacity of fish blood,resulting in damage to multiple organs.At present,research on the effect of nitrite on fish is concentrated on acute stress,such as Takifugu rubripes,GIFT Oreochromis niloticus and Cyprinus carpio.However,there is no report on the study of nitrite on chronic stress of Acipenser sinensis.To evaluate the effects of chronic nitrite stress on growth,antioxidative and immunity indices of Acipenser sinensis,the fish were exposed to different nitrite levels(0.01,0.20,0.40,0.80,1.60 mg·L-1)for 60 days.Then,the paramaters including growth,antioxidative and immunity indices were measured.Results showed that the death rate of F2 Acipenser sinensis in the high-concentration group was 33.33%,and the survival rate of all other groups was 100%.The weight gain rate(WGR)and specific growth rate(SGR)of F2 Acipenser sinensis gradually decreased with the increase of nitrite concentration(P<0.05),but there was no significant change in viserosomatic index(VSI),hepatosomatic index(HSI)and condition factor(CF).In the antioxidative indexes,the superoxide dismutase(SOD),catalase(CAT),total antioxidant capacity(T-AOC)activity and glutathione(GSH)content in serum and liver gradually decreased with the increase of nitrite concentration,while MDA content increased.In the immune indexes,the LZM activity and IgM content in serum and liver decreased significantly with the increase of nitrite concentration(P<0.05).These results suggested that the chronic nitrite stress significantly inhibited the growth of the fish,and reduced the antioxidative and immunity capacity of F2 Chinse sturgeon.This had important reference for understanding the nitrite stress response of Acipenser sinensis and its healthy breeding.
