摘要
为探讨白芷乳剂对过敏性鼻炎小鼠血清细胞因子及鼻黏膜中Th17/Treg平衡的影响,采用卵清蛋白诱发过敏性鼻炎小鼠模型,将C57BL/6小鼠随机分为正常组、模型组和白芷组。采用苏木精-伊红(hematoxylin-eosin,HE)染色观察鼻黏膜组织的病理变化,FCM检测血液中Th17和Treg的比例,ELISA检测血清和鼻腔灌洗液中IgE、IL-6、IL-10、IL-17和转化生长因子β1(transforming growth factorβ1,TGF-β1)的水平,RT-PCR检测鼻黏膜中IL-17和TGF-β1 mRNA的表达。HE染色结果显示:与正常组比较,模型组有大量嗜酸性粒细胞为主的炎症细胞浸润;与模型组比较,白芷组鼻黏膜上皮细胞排列整齐,上皮破损状况良好,嗜酸性粒细胞浸润相对减少。与正常组比较,模型组Th17比例增多(P<0.01),Treg比例减少(P<0.05),血清和鼻腔灌洗液中IL-6和IL-17水平显著升高(P<0.01),IL-10和TGF-β1水平显著降低(P<0.01);与模型组比较,白芷组Th17比例减少(P<0.01),Treg比例增多(P<0.05),血清和鼻腔灌洗液中IL-6和IL-17水平显著降低(P<0.01),IL-10和TGF-β1水平显著升高(P<0.05)。这提示白芷能有效缓解过敏性鼻炎小鼠的症状,减轻其鼻黏膜组织损伤,其机制可能与下调Th17比例,上调Treg比例,恢复Th17/Treg平衡有关。
we aimed to investigate the effect of radix angelicae emulsion on serum cytokines and Th17/Treg balance in nasal mucosa of mice with allergic rhinitis. C57 BL/6 mice were randomly divided into normal group, model group and radix angelicae group. The model of allergic rhinitis was induced by ovalbumin. The pathological changes of nasal mucosa were observed by hematoxylin-eosin(HE) staining. The ratio of Th17 and Treg in peripheral blood was detected by flow cytometry. The levels of IgE, IL-6, IL-10, IL-17 and transforming growth factor β1(TGF-β1) in serum and nasal lavage fluid were detected by ELISA. The expressions of IL-17 and TGF-β1 mRNA in nasal mucosa lavage fluid were detected by RT-PCR. The HE staining results showed that, compared with the normal group, the model group had a large number of eosinophil-based inflammatory cell infiltration;and compared with the model group, the nasal mucosa epithelial cells in the radix angelicae group were neatly arranged, the epithelial damage was slight, and the eosinophil infiltration was relatively cut back. Compared with the normal group, the proportion of Th17 in the model group increased(P<0.01)whereas the proportion of Treg decreased(P<0.01), and the levels of IL-6 and IL-17 in serum and nasal lavage fluid increased significantly(P<0.01), and the levels of IL-10 and TGF-β1 decreased significantly(P<0.01). Compared with the model group, the proportion of Th17 in the radix angelicae group decreased(P<0.01), the proportion of Treg increased(P<0.05), the levels of IL-6 and IL-17 in serum and nasal lavage fluid were significantly decreased(P<0.01), and the levels of IL-10 and TGF-β1 were significantly increased(P<0.05). In summary, radix angelicae can alleviate the nasal mucosa damage of allergic rhinitis mice, probably by restoring Th17/Treg balance.
