摘要
目的建立山楂饮片及炮制品的HPLC指纹图谱。方法采用HPLC法,分别测定山楂饮片、焦山楂及山楂炭醇提取物的指纹图谱,色谱条件:Kromasil色谱柱(250 mm×4.6 mm,5μm);流动相为乙腈(A)-0.1%乙酸水溶液(B)梯度洗脱;检测波长为325 nm;柱温为30℃;流速为1 mL/min;进样量为20μL。结果11批山楂饮片中均标示出14个特征峰,山楂焦品共标示出12个特征峰,山楂炭品共标示出10个特征峰。通过对色谱峰的分析,山楂焦品和山楂炭品均检测出山楂饮片中的8个共有峰;指认了其中3个特征峰,分别为5-羟甲基糠醛、绿原酸、金丝桃苷。结论山楂饮片炮制前后的指纹图谱存在显著差异,方法准确可靠,可为山楂饮片及其炮制品质量控制提供参考依据。
Objective To establish high performance liquid chromatography(HPLC)fingerprint of Hawthorn pieces,charred Hawthorn and Hawthorn charcoal.Methods The fingerprint of alcohol extracts from hawthorn decoction pieces,charred hawthorn and hawthorn charcoal was determined by reversed-phase HPLC.The chromatographic conditions were as follows:Kromasil chromatographic column(250 mm×4.6 mm,5μm),Acetonitrile(A)and 0.1%Acetic acid aqueous solution(B)as a mobile phase in gradient elution,the detection wavelength at 325 nm,column temperature at 30℃,the flow rate of 1 mL/min and the injection volume of 20μL.Results 14 characteristic peaks were identified in 11 batches of Hawthorn pieces.Charred Hawthorn was marked with 12 characteristic peaks.Hawthorn charcoal was marked with 10 characteristic peaks.According to the analysis of chromatographic peaks,Charred Hawthorn and Hawthorn charcoal were both detected 8 common peaks in Hawthorn pieces.In addition,3 characteristic peaks were marked,namely 5-hydroxymethylfurfural,Chlorogenic acid and Hyperoside.Conclusion There are significant differences in fingerprint of Hawthorn pieces before and after processing.The method is accurate,reliable,which can provide reference for the quality control formulation of Hawthorn pieces and hawthorn processed products.
