摘要
目的研究大黄提取物对脑缺血/再灌注大鼠脑组织中凋亡相关因子的影响,进一步探讨大黄提取物对缺血性脑损伤大鼠的作用机制。方法成年♂SD大鼠45只,随机分成3组:假手术组(sham)、模型组(MCAO)和大黄提取物组(MCAO+rhubarb extract),大黄提取物给药剂量为200 mg·kg^-1,大鼠大脑中动脉栓塞模型(middle cerebral artery occlusion,MCAO)制备后,连续灌胃给药6 d。TUNEL法检测细胞凋亡,免疫荧光法观察大鼠脑组织中GFAP表达,RT-qPCR检测IL^-1β、iNOS mRNA表达,Western blot检测GFAP、Bax、Bcl-2、cleaved caspase-3和caspase-3蛋白表达。结果与模型组相比,大黄提取物能降低TUNEL阳性细胞和GFAP表达,降低IL^-1β、iNOS mRNA表达,降低GFAP、Bax、cleaved caspase-3蛋白表达,升高Bcl-2蛋白表达。结论大黄提取物对缺血性脑损伤大鼠具有神经保护作用,与降低GFAP、Bax、cleaved caspase-3蛋白,提高Bcl-2蛋白表达,抑制脑组织细胞凋亡有关。
Aim To study the effect of rhubarb extract on apoptosis related factors in brain tissues of rats with cerebral ischemia reperfusion,and further explore the mechanism of rhubarb extract on rats with ischemic brain injury.Methods Forty-five adult male Sprague-Dawley rats were randomly divided into three groups:sham operation group(sham),model group and rhubarb extract group.Rat middle cerebral artery occlusion model(MCAO)was prepared by suture method.The rhubarb extract was administered at a dose of 200 mg·kg^-1,and administered continuously for 6 days after preparation of MCAO model.Apoptosis was detected by TUNEL method.The expression of GFAP in rat brain tissue was observed by immunofluorescence.The expression of IL^-1βand iNOS mRNA was detected by RT-qPCR.GFAP,Bax,Bcl-2,cleaved caspase-3 and caspase-3 were detected by Western blot.Results Compared with model group,rhubarb extract decreased the expression of TUNEL positive cells and GFAP,decreased the expression of IL^-1βand iNOS mRNA,decreased the expression of GFAP,Bax,cleaved caspase-3 protein,and increased the expression of Bcl-2.Conclusions Rhubarb extract has neuroprotective effect on rats with ischemic brain injury,which is related to the decrease of GFAP,Bax,cleaved caspase-3 protein expression,increase of Bcl-2 protein expression and inhibition of brain tissue apoptosis.
