芍药甘草汤通过抑制NF-κB信号通路减轻兔颈椎间盘炎症损伤的作用及机制研究 被引量:24

Study on the effects and mechanism of Shaoyao Gancao Decoction in reducing cervical intervertebral disc inflammation injury in rabbits by inhibiting NF-κB signaling pathway

摘要 目的:探讨芍药甘草汤对兔颈椎间盘炎症损伤的作用及其可能的机制。方法:将6月龄的新西兰白兔随机分为高、中、低剂量组,模型组,正常组,每组6只,除正常组外其余各组参照寒湿型兔颈型颈椎病模型进行造模。造模8周后,高、中、低剂量组分别予以16、8、4g/kg的芍药甘草汤进行灌胃治疗,正常组和模型组给予等量的0.9%氯化钠溶液。于治疗4周后用ELISA法检测各组血清中炎症因子IL-1β、IL-6、TNF-α的表达水平。HE染色法观察各组兔颈椎间盘组织形态的变化。Western Blot法检测各组兔颈椎间盘组织中NF-κB信号通路关键节点髓样分化因子88(MyD88)、核转录因子(NF)-κB P65蛋白表达水平。结果:与模型组比较,芍药甘草汤可降低兔血清炎症因子IL-1β、IL-6、TNF-α的表达(P<0.01);能明显改善受炎症损伤的兔颈椎间盘细胞形态结构;同时可降低MyD88、NF-κB P65蛋白的表达(P<0.01)。结论:芍药甘草汤可以减轻兔颈椎间盘的炎症损伤,其作用机制可能是抑制NF-κB信号通路释放IL-1β、IL-6、TNF-α等炎症因子。 Objective:To investigate the effects of Shaoyao Gancao Decoction on cervical intervertebral disc inflammation injury in rabbits and explore its possible mechanism.Methods:Six-month-old New Zealand white rabbits were randomly divided into high-dose group,medium-dose group,low-dose group,model group and normal group(n=6).The model was established according to the model of cold-wet rabbit neck cervical spondylosis.Eight weeks after modeling,high-dose,medium-dose,and low-dose groups were given gavage of 16,8,4 g/kg Shaoyao Gancao Decoction,and the normal group and the model group were fed normally.The levels of IL-1β,IL-6 and TNF-αin serum were detected by ELISA after 4 weeks of treatment.The changes of cervical disc morphology were observed by HE staining.Western Blot was used to detect the protein expression of myeloid differentiation factor 88(MyD88)and nuclear factor(NF)-κB P65 in the key nodes of NF-κB signaling pathway in the rabbit cervical intervertebral disc tissue.Results:Shaoyao Gancao Decoction could decrease the expressions of IL-1β,IL-6 and TNF-αin rabbit serum(P<0.01).It could significantly improve the morphological structure of cervical intervertebral disc cells injured by inflammation.At the same time,it could reduce the expressions of MyD88 and NF-κB P65(P<0.01).Conclusion:Shaoyao Gancao Decoction can reduce the inflammatory injury of rabbit intervertebral discs,and its mechanism may be to inhibit the NF-κB signaling pathway releasing IL-1β,IL-6,TNF-αand other inflammatory factors.
