摘要
为了探讨黑龙江某规模化牧场发病奶牛的致病原及其生物学特征,对黑龙江省某规模化牛场采集到的育成牛肺脏组织样本进行病毒分离鉴定,确定了样本的基因型为BVDV-1d,5’UTR的同源性比对显示,与中国北京株BJ1023(Genbank:KF925509)同源性最高。E2基因的同源性比对显示,与中国北京株BJ1308(Genbank:KT951841)同源性最高。对样本进行E2基因序列分析和理化性质分析可知,E2核苷酸水平上有58.9%的可变性位点(763/1296),保守区有两段:核苷酸位置394~541以及919~1234。有两个潜在的跨膜信号区域:30~50和410~420。蛋白不稳定性为33.62,可以将该蛋白质分类为稳定的蛋白质。E2基因的抗原表位与瑞士毒株SuwaCp(Genbank:KC853441)的抗原表位优势部分走势基本一致。分离获得的毒株为我国BVDV毒株资源与分子流行病学提供了资料。
In order to investigate the pathogenic factors and biological characteristics of pathogenic cows in a large-scale farm in Heilongjiang province,virus isolation and identification were carried out on the lung tissue samples,the genotype of the samples was determined to be bvdv-1d.The homology comparison of 5’UTR showed the highest homology with BJ1023(Genbank:KF925509)from Beijing,China.The homology comparison of E2 gene showed the highest homology with Beijing strain BJ1308(Genbank:KT951841).Analysis of E2 gene sequence and physicochemical properties of the samples showed that 58.9%of the E2 nucleotide was variable(763/1296),and there were two conserved regions:nucleotide position 394~541 and 919~1234.Six potential glycation sites were found(31NMSN,96NLTE,176NKTQ,260NGTV,330NFSR,369NLTS),with two potential transmembrane signal regions:30~50 and 410~420.The protein instability was 33.62,which could be classified as stable protein.The epitope of E2 protein showed the same trend as that of SuwaCp(Genbank:KC853441).The isolated strains provided data for the resources and molecular epidemiology of BVDV strains in China.
