摘要
骨骼肌快肌肌钙蛋白(Troponin C Fast Skeletal Muscle,TNNC2)是骨骼肌快肌收缩过程中重要的调控蛋白,本研究利用荧光定量PCR技术较为系统地检测了TNNC2基因在新生秦川牛和成年秦川牛心脏、肝脏、脾脏、肺脏、肾脏、大肠、小肠、瘤胃、网胃、瓣胃、皱胃、肾周脂肪、皮下脂肪、背最长肌等13种组织中的表达特性,同时分析了其在体外培养的牛骨骼肌成肌细胞不同分化时期的表达规律。结果发现:TNNC2基因在牛快肌组织(背最长肌、后腿肌等)中表达量显著高于其他组织(心脏、肝脏、脾脏、肺脏等),在成年牛背最长肌中的表达水平显著高于新生牛,而成年牛和新生牛在脾脏和瘤胃中的表达水平无明显差异;随着成肌细胞的分化,TNNC2基因在成肌细胞分化过程中的表达水平呈上升趋势。综上,TNNC2基因对肉牛肌肉组织的生长发育可能具有重要的调控作用,可对其进行深入的功能研究以期用于肉牛的分子育种实践中。
Skeletal muscle troponin(Troponin C,skeletal muscle,TNNC2)is an important regulatory protein in skeletal muscle fast muscle contraction.In this study,the expression characteristics of TNNC2 in 13 tissues of newborn and adult Qinchuan cattle were systematically detected using real-time quantitative PCR technology,and its expression level in different periods of differentiation of bovine skeletal muscle myoblasts cultured in vitro were further analyzed.Results showed that the expression level of TNNC2 gene in bovine fast muscle tissue(longest dorsal muscle,hind leg muscle,etc.)is significantly higher than that in other tissues(heart,liver,spleen,lung,etc.).The expression level in the longest muscle of bovine dorsal is significantly higher than that in newborn cattle,and there is no significant difference in the expression level of spleen and rumen between adult cattle and newborn cattle.With the differentiation of myoblasts,the expression level of this gene is increasing.The characteristics of TNNC2 gene expression reveals this gene may have important regulatory effects on the growth and development of beef muscle tissue,and it can be studied in depth for its functional application in the molecular breeding practice of beef cattle.
