摘要
目的比较不同年龄患者胚胎氨基酸代谢之间的差异,揭示不同年龄患者胚胎的氨基酸代谢规律,为改善胚胎培养环境、获得更优质胚胎从而提高体外受精-胚胎移植(IVF-ET)的成功率提供依据。方法选择接受IVF-ET治疗的患者,收集培养过胚胎的废弃培养液微滴,按女方患者的年龄分为<35岁组、35~39岁组、≥40岁组,每年龄组各收集20份培养液标本,共60份标本,另外选择同一时期同样培养环境下未培养过胚胎的培养液为空白对照组。采用高效液相色谱法(HPLC)检测标本中20种游离氨基酸的含量,比较不同年龄组培养液中氨基酸含量的差异。结果3组胚胎培养液中天冬氨酸、天冬酰胺、丝氨酸、谷氨酰胺、甘氨酸、苏氨酸、组氨酸、丙氨酸、牛磺酸、丙氨酰谷氨酰胺、酪氨酸、缬氨酸、蛋氨酸、色氨酸、异亮氨酸、亮氨酸以及赖氨酸含量比较差异有统计学意义(P均<0.05)。<35岁组胚胎培养液中氨基酸减少量低于≥40岁组。<35岁组胚胎培养液中氨基酸转换量低于≥40岁组。结论患者的年龄不同,其胚胎的氨基酸代谢是有差异的,患者的年龄越大,其胚胎的氨基酸减少量和氨基酸转换量越大。
Objective To compare the differences of amino acid metabolism and to reveal the patterns of amino acid metabolism among women of different ages undergoing in vitro fertilization and embryo transfer(IVF-ET),aiming to provide basis for improving the embryo culture environment,obtaining higher quanlity embryos and enhancing the success rate of IVF-ET.Methods The waste culture medium droplets of cultured embryos were collected from patients receiving IVF-ET.According to the age of female patients,the samples were divided into three groups(<35 years old,35-39 years old,and≥40 years old).Twenty samples were collected in each age group.The culture medium without embryo culture under the same culture environment in the same period was utilized as the empty control.The concentration of 20 free amino acids in the samples was detected by high performance liquid chromatography(HPLC).The differences of amino acid concentration were statistically compared among three groups.Results The concentration of aspartic acid,asparagine,serine,glutamine,glycine,threonine,histidine,alanine,taurine,alanyl glutamine,tyrosine,valine,methionine,tryptophan,isoleucine,leucine and lysine significantly differed in the embryo culture media among three groups(all P<0.05).The decrease of amino acids in the embryo culture medium in the<35 years old group was lower than that in the≥40 years old group.The amount of amino acid turnover in the embryo culture medium in the<35 years old group was lower compared with that in the≥40 years old group.Conclusions The amino acid metabolism significantly differs among IVF-ET patients of different ages.The older the patients,the greater the amount of amino acid reduction and amino acid turnover in the embryos.
