超高效液相色谱法同时测定清宣止咳颗粒中5种有效成分的含量 被引量:5

摘要 目的建立超高效液相色谱(UPLC)法同时测定清宣止咳颗粒中苦杏仁苷、芍药苷、芦丁、柚皮苷和橙皮苷5种有效成分的含量。方法采用Waters HSS T3 C 18色谱柱(100 mm×2.1 mm,1.8μm);流动相为乙腈-0.05 mol·L^-1磷酸二氢钾;梯度洗脱;流速:0.21 mL·min^-1;检测波长:230 nm;柱温:35℃。结果苦杏仁苷、芍药苷、芦丁、柚皮苷和橙皮苷分别在0.0118~0.1184(r 1=0.9997),0.0091~0.0910(r 2=0.9997),0.0101~0.1012(r 3=0.9998),0.0151~0.1512(r 4=0.9998)和0.0119~0.1191μg(r 5=0.9996)范围内与峰面积线性关系良好,平均回收率(n=6)均在97.70%~101.68%范围内,相应的RSD值均在1.26%~1.96%范围内。结论该方法简便、快速、灵敏度高、重复性好,可用于清宣止咳颗粒中苦杏仁苷、芍药苷、芦丁、柚皮苷和橙皮苷5种有效成分的含量测定。 Objective To established a ultra performance liquid chromatography(UPLC)method for simultaneous determination of 5 active ingredients,amygdalin,paeoniflorin,rutin,naringin and hesperidin in Qingxuanzhike Granules.Methods The determination was performed on Waters HSS T3 C 18 column(100 mm×2.1 mm,1.8μm)with mobile phase consisted of acetonitrile-0.05 mol·L^-1 potassium dihydrogen phosphate in a gradient elution at a flow rate of 0.21 mL·min^-1.The UV detection wavelength was set at 230 nm and the column temperature was set at 35℃.Results Amygdalin,paeoniflorin,rutin,naringin and hesperidin showed good linear relationship with the peak area in the ranges of 0.0118-0.1184(r 1=0.9997),0.0091-0.0910(r 2=0.9997),0.0101-0.1012(r 3=0.9998),0.0151-0.1512(r 4=0.9998)and 0.0119-0.1191μg(r 5=0.9996)respectively.The average recovery rates(n=6)were 97.70%-101.68%with corresponding RSD of 1.26%-1.96%.Conclusion The method is simple,fast,sensitive and reproducible,and can be used for simultaneous determination of paeoniflorin,amygdalin,rutin,naringin and hesperidin in Qingxuanzhike Granules.
