摘要
为了构建菊芋(Helianthus tuberosus)种质的耐寒性体系,以4个菊芋品种为实验材料,通过人工模拟低温的方法测定了菊芋块茎的丙二醛(MDA)、可溶性糖(SS)和脯氨酸(PRO)含量以及相对电导率(REC)等生化指标,并测定了低温处理后块茎的生长发育指标。利用相对电导率结合Logistic方程计算菊芋块茎的半致死温度(LT50),结合隶属函数法对菊芋耐寒性进行了综合评价。结果表明,4种参试材料在-30℃处理6 h时,块茎存活率呈现显著差异,其中‘青芋1号’与‘青芋4号’在该处理下全部死亡,‘青芋2号’与‘青芋3号’的存活率分别达到了70%与60%;适当的低温和适当的处理时间可以提高发芽率;与对照相比,低温胁迫下的SS、MDA、PRO含量随着胁迫时间增加呈现逐渐升高的趋势;4个菊芋品种在各个低温处理下的LT50值分布在-23.7^-2.4℃;根据综合隶属值分析可知4个菊芋品种的耐寒性强弱为:‘青芋2号’>‘青芋1号’>‘青芋4号’>‘青芋3号’。该耐寒性评价体系可广泛用于不同菊芋种质耐寒性的快速鉴定和预测。
The present study aimed to establish a system for evaluating chilling tolerance of Jerusalem artichoke(Helianthus tuberosus)germplasm variety,the contents of malondialdehyde(MDA),soluble sugar(SS)and proline(PRO)and relative conductivity(REC)of Jerusalem artichoke tubers,and the growth and development indexes were determined by artificial simulation of low temperature.The semi-lethal temperature(LT50)of Jerusalem artichoke tuber was calculated by using relative conductivity combined with Logistic equation,and the comprehensive evaluation method of Jerusalem artichoke cold resistance was established by combining membership function method.The results showed that the survival rate of tuber showed significant difference during 6 h of–30℃treatment.Among them,no tuber was survival of‘Qingyu No.1’and‘Qingyu No.4’,however,the survival rates of‘Qingyu No.2’and‘Qingyu No.3’were 70%and 60%respectively.Appropriately low temperature and treatment time promoted the germination rate.The content of SS,MDA,PRO under low temperature stress increased gradually with the increase of stress time compared with the control.The LT50 values of 4 varieties were range from-23.7–-2.4℃under various low temperature treatments.The cold resistance of four varieties of Jerusalem artichoke was as follows:‘Qingyu No.2’>‘Qingyu No.1’>‘Qingyu No.4’>‘Qingyu No.3’according to the comprehensive membership function evaluation.This cold tolerance evaluation system can be widely used for rapid identification and prediction of cold tolerance in different Jerusalem artichoke germplasm.
