针刺捻转补泻手法对自发性高血压大鼠顶叶皮质IL-6、TNF-α及NGF影响的研究 被引量:11

摘要 目的通过观察针刺捻转补泻手法对自发性高血压(SHR)大鼠顶叶皮质中白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)及神经生长因子(NGF)表达水平的影响,探讨高血压脑损伤过程中的炎症反应机制及针刺的作用机理。方法选用64只9周龄雄性SHR大鼠,随机分为模型组、捻转补法组、捻转泻法组、电针组,每组16只,以及16只Wistar-Kyoto(WKY)大鼠为空白组。模型组和空白组只抓取固定,每日抓取固定1次,共干预14 d。其余3组针刺组分别采用针刺捻转补法、针刺捻转泻法及电针刺激,每日1次,共干预14 d。各组大鼠于针刺前1 d、针刺第2、6、10、14天,用恒温式无创血压仪观察各组大鼠尾压。采用ELISA技术检测大鼠顶叶皮质中IL-6、TNF-α和NGF的表达水平。结果模型组大鼠收缩压相比空白组明显升高(P<0.01);捻转泻法组、捻转补法组、电针组大鼠收缩压相比模型组均明显下降(P<0.01),且捻转泻法组降压效果最快,降压幅度最大;针刺后,捻转泻法组、捻转补法组、电针组大鼠顶叶皮质中的IL-6、TNF-α水平相比模型组明显下降(P<0.05),NGF的含量相比模型组明显上升(P<0.01)。结论针刺能够降低SHR大鼠收缩压,降低顶叶皮质中的IL-6、TNF-α含量,抑制神经炎症反应,同时增加顶叶皮质中NGF的含量,促进神经元的修复,缓解高血压脑损伤。针刺降压的中枢机制可能与抑制神经炎症反应,同时促进神经元的修复密切相关。 Objective:To observe the effect of reinforcing and reducing methods by twirling and rotating the needle on contents of IL-6,TNF-αand NGF in parietal cortex of spontaneously hypertensive rats,and to explore the mechanism of inflammatory reaction and acupuncture in hypertensive brain injury.Methods:649-week-old male SHR were randomly divided into the model group,acupuncture with reinforcing method group,acupuncture with reducing method group,electroacupuncture group,16 rats in each,16 WKY rats as the control group.The model group and the control group were only fixed for grasping once a day for 14 days.The other three groups were treated with acupuncture with reinforcing method,acupuncture with reducing method and electroacupuncture stimulation once a day for 14 days.Rats were observed with constant temperature noninvasive blood pressure meter on the day before acupuncture and on the 2 nd,6 th,10 th and 14 th days after acupuncture.The expression levels of IL-6,TNF-αand NGF in parietal cortex of rats were detected by ELISA.Results:Systolic blood pressure in the model group was significantly higher than that in the control group(P<0.01).Compared with the model group,the systolic blood pressure in acupuncture with reinforcing method group,acupuncture with reducing method group and electroacupuncture group was obviously reduced(P<0.01),and acupuncture with reducing method group had the fastest anti-hypertensive effect and the largest anti-hypertensive amplitude.After acupuncture,the levels of IL-6 and TNF-αin parietal cortex of rats in acupuncture with reinforcing method group,acupuncture with reducing method group and electroacupuncture group were significantly lower than those in the model group(P<0.05),and the content of NGF was significantly higher than that in the model group(P<0.01).Conclusion:Acupuncture can reduce systolic blood pressure of SHR,reduce IL-6 and TNF-αcontent in parietal cortex,inhibit neuroinflammatory reaction,increase NGF content in parietal cortex,promote neuronal repair,and relieve hypertensive brain injury.The central mechanism of acupuncture hypotension may be closely related to the inhibition of neuroinflammatory reaction and the promotion of neuronal repair.
