不同因素对新型冠状病毒核酸检测结果的影响分析 被引量:6

摘要 目的研究不同因素对新型冠状病毒(COVID-19)核酸检测结果的影响。方法以2 122例送检的标本为研究对象,通过样本采集人员及检测人员培训前后结果比对、样本不同采集部位、同一厂家不同核酸提取方法、不同生产厂家核酸检测试剂等方面进行研究,验证不同因素对新型冠状病毒核酸检测的影响。结果样本采集人员检测结果的Ct值和ΔRn值差异均有统计学意义(P<0.05);实验室检测人员培训前后检测结果Ct值和ΔRn值差异均有统计学意义(P<0.05);鼻咽拭子的混合样本与咽拭子样本检测结果的Ct值和ΔRn值差异有统计学意义(P<0.05);不同核酸提取方法检测结果的Ct值和ΔRn值差异有统计学意义(P<0.05),核酸A260/280比值差异有统计学意义(P<0.05)。6种国产试剂均能检出阳性标本,但将阳性标本进行1∶256倍稀释后E检测试剂仍能检出阳性标本。结论分析新型冠状病毒核酸检测前、中2个环节中的多种影响因素,防止假阴性、假阳性等错误报告的产生。 Objective To identify and investigate the influencing factors for nucleic acid detection of Covid-19.Methods 2122 submitted specimens were collected.The results done by the collectors and detection personnel before training and after training,the difference of specimen sites,the different extraction method using the same manufacturer’s amplification reagents and also adoption of different manufacture’s detection platforms were compared.Results The difference of CT value andΔRN value of the test results of sample collection personnel was statistically significant(P<0.05),the difference of CT value andΔRN value of the test results before and after the training of laboratory testing personnel was statistically significant(P<0.05).The difference of CT value andΔRN value of mixed samples of nasopharyngeal swab and throat swab samples was statistically significant(P<0.05).The Ct value andΔRn value showed statistically significant difference using different extraction methods(P<0.05).All 6 domestic platforms detected positive specimens,but only E platform detected positive specimens at 1:256 times dilution.Conclusion multiple factors influencing the Covid-19 detection throughout the entire procedures may be compared and analyzed in order to prevent false negative,false positive and other false reports.
