糜子幼胚愈伤组织的诱导及植株再生

摘要 为研究影响糜子幼胚愈伤组织诱导分化的因素,优化培养条件,提高糜子幼胚培养效率。以‘陕糜1号’和‘陕糜2号’为材料,研究低温预处理时间、激素种类及质量浓度对糜子幼胚愈伤组织诱导、分化和再生的影响。结果表明,4℃低温预处理3 d能显著提高糜子的幼胚愈伤组织诱导率;2种糜子最适宜的诱导培养基均为MS+3.0 mg/L 2,4-D。添加一定质量浓度的KT,可以改善愈伤组织质量,但是2种糜子的愈伤组织诱导率有显著的基因型差异。可见,2种糜子最适宜的分化培养基均为MS+1.0 mg/L 6-BA+0.1 mg/L NAA,且绿点率均在80%以上。 This study is to optimize the culture conditions of callus induction and differentiation for improving the culture efficiency of proso millet immature embryos.With Shaanmi No.1(SM1)and Shaanmi No.2(SM2)as experimental materials,the effects of low temperature,type and concentrations of hormones on induction,differentiation and regeneration of calli from immature embryos of proso millet were studied.The results showed that pretreatment of 4℃low temperature for 3 days significantly increased frequency of callus induction from proso millet immature embryos.The suitable callus induction medium for two proso millet varieties was MS medium with 2,4-D(3.0 mg/L).Addition of a certain concentration of kinetin(KT)could improve callus quality,and callus induction frequency showed significantly genotype differences between the two varieties.The most suitable differentiation medium for both proso millets was MS+1.0 mg/L 6-BA+0.1 mg/L NAA(0.1 mg/L),and the green spot rate was above 80%.
