摘要
目的探讨长链非编码RNA(long non-coding RNA,lncRNA)ZFAS1(lncZFAS1)对食管癌细胞增殖、凋亡及迁移的影响,并初步探讨其机制。方法采用qRT-PCR法检测食管癌组织及食管癌细胞中lncZFAS1的表达;shRNA干扰lncZFAS1表达后,采用CCK-8法、流式细胞术、划痕试验、Transwell试验检测其对细胞增殖、凋亡、迁移及侵袭的影响;建立小鼠移植瘤模型,检测敲低lncZFAS1对移植瘤生长的影响。结果lncZFAS1在食管癌组织及食管癌细胞中的表达均明显高于癌旁组织及正常食管上皮细胞。敲低lncZFAS1可显著抑制食管癌细胞的增殖、迁移及侵袭能力;可使食管癌细胞中miR-150-5P表达上调,HMGA2表达下调;可显著抑制裸鼠移植瘤的生长。结论lncZFAS1可促进食管癌细胞的增殖及迁移,其机制可能与调控miR-150-5P/HMGA2分子轴相关。
Objective To investigate the effect of long non-coding RNA ZFAS1(lncZFAS1)on proliferation,apoptosis and migration of esophageal cancer cells as well as relevant mechanism.Methods The expressions of lncZFAS1 in esophageal cancer tissue and cells were determined by qRT-PCR.The influences of interference of lncZFAS1 expression by shRNA on proliferation,apoptosis,migration and invasion of esophageal cancer cells were evaluated by CCK-8 method,flow cytometry,scratch test and Transwell assay respectively.Mouse model of transplanted tumor was established to evaluate the effect of knock-down of incZFAS1 expression on growth of transplanted tumor.Results Both the expression levels of lncZFAS1 in esophageal cancer tissue and cells were significantly higher than those in paracancerous tissue and normal esophageal epithelium.The knock-down of lncZFAS1 expression inhibited the proliferation,migration and invasion abilities of esophageal cancer cells significantly,while up-regulated the miR-150-5 P and down-regulated the HMGA2 expression in esophageal cancer cells and inhibited the growth of transplanted tumor in nude mice significantly.Conclusion The lncZFAS1 promoted the proliferation and migration of esophageal cancer cells significantly,of which the mechanism might be associated with the regulation of molecular axis of miR-150-5 P/HMGA2.
