摘要
目的分析肾透明细胞癌(ccRCC)基因表达变化及预后相关因素。方法运用R/Bioconductor分析平台,基于肿瘤基因组图谱计划(TCGA)中的ccRCC研究数据,运用Limma算法筛选肿瘤组与对照组间倍数改变>2,P<0.05的基因为差异表达基因。进一步以患者年龄、性别、TNM分期、基因表达值为影响因素分析并绘制Kaplan-Meier生存曲线。结果在肿瘤组织中筛选出4837个差异表达基因(上调基因1974个,下调基因2863个),其中252个差异基因倍数改变>5个(27个上调基因,225个下调基因);性别因素对患者整体生存率无显著影响(P>0.05);随着年龄增加以及TNM分期的进展,患者整体生存率下降(P<0.05);随着基因NPTX2、HSF4表达升高,患者整体生存率下降(P<0.05);随着基因TMEM213表达升高,患者整体生存率上升(P<0.05)。结论本研究鉴定了肾透明细胞癌的基因表达变化及预后相关因素,能够为疾病的分子诊断与预后评估等提供依据。
Objective To analyze the gene expression patterns and the prognosis-related factors of clear cell renal cell carcinoma(ccRCC).Methods The R/Bioconductor platform was applied for data processing and analysis based on the ccRCC data in The Cancer Genome Atlas(TCGA)project,genes with fold change(FC)>2 and P<0.05 analyzed by the limma algorithm between tumors and normal controls were selected as differentially expressed genes(DEGs).Kaplan-Meier survival curves based on age,gender,TNM stage,and the expression values of DEGs were analyzed.Results There were 4837 DEGs(FC>2,1974 up-regulated genes and 2863 down-regulated genes)in tumors compared with normal controls.Among them,there were 252 DEGs with FC>5 in the tumor tissues(27 up-regulated genes and 225 down-regulated genes).The factor of gender had no significant effect on the overall survival rate of patients(P>0.05).With the increase of age and the progress of TNM stage,the overall survival rate of patients decreased(P<0.05).As the expression of genes NPTX2 and HSF4 increased,the overall survival rate of patients decreased(P<0.05).As the expression of gene TMEM213 increased,the overall survival rate of patients increased(P<0.05).Conclusion The study analyzed and identified the gene expression changes and prognostic-related factors of ccRCC,which can provide a basis for the molecular diagnosis and prognosis evaluation of the disease.
