摘要
目的:探讨C20orf54在贲门腺癌(GCA)组织中的表达和意义。方法:采用RT-PCR、Western blot和免疫组化SP法检测60例GCA组织及癌旁正常黏膜组织中C20orf54的表达。对C20orf54高、低表达的GCA患者进行生存分析比较。结果:GCA组织中C20orf54 mRNA及蛋白的表达低于癌旁正常黏膜组织(P<0.001);GCA和癌旁正常黏膜组织中C20orf54蛋白的高表达率,差异有统计学意义(P<0.001)。中、低分化,有淋巴结转移,浸润深度T3、T4和临床分期Ⅲ、Ⅳa期的GCA组织中C20orf54蛋白高表达率低于高分化,无淋巴结转移,浸润深度T1、T2和临床分期Ⅰ、Ⅱ期者(P<0.05)。C20orf54高表达和低表达GCA患者Kaplan-Meier生存曲线差异有统计学意义(P<0.001),中位生存期分别是40.2和23.1个月。结论:C20orf54表达下调可能与GCA发生发展、侵袭及不良预后有关。
Aim:To detect the expression of C20orf54 in gastric cardia adenocarcinoma(GCA)tissue.Methods:A total of 60 primary GCA specimens and adjacent normal mucosa tissue were collected to examine the mRNA and protein expressions of C20orf54 by RT-PCR,Western blot and immunohistochemistry.Kaplan-Meier survival analysis was conducted between patients with high and low expression of C20orf54.Results:C20orf54 mRNA and protein expressions in GCA tissue were significantly lower than those in adjacent normal mucosa tissue(P<0.001).C20orf54 high expression rate in GCA tissue and adjacent normal mucosa tissue had significant difference(P<0.001).C20orf54 expression was lower in tissues with infiltration degree T3,T4,median and low degree of differentiation,lymph node metastasis and clinical stageⅢ,Ⅳa than those in tissues with infiltration degree T1,T2,high degree of differentiation,no lymph node metastasis and clinical stageⅠ,Ⅱ(P<0.05).The Kaplan-Meier survival curves of patients with C20orf54 high expression and low expression had significant difference(P<0.001),and the median overall survival was 40.2 months and 23.1 months,respectively.Conclusion:C20orf54 low expression may be associated with carcinogenesis,development,invasive ability and poor prognosis of GCA.
