沉默叉头框转录因子C1基因对肺腺癌A549细胞迁移和侵袭能力的影响 被引量:1

Influence of forkhead box C1 silencing on migration and invasion ability of lung adenocarcinoma A549 cells

摘要 目的:探讨下调叉头框转录因子C1(FOXC1)基因的表达对肺腺癌A549细胞迁移及侵袭能力的影响。方法:实验分为3组,空白对照组(正常培养A549细胞),siRNA-FOXC1转染组和siRNA-NC转染组。转染6 h后,应用实时荧光定量PCR法和Western blot法检测细胞中FOXC1、E-cadherin、Vimentin mRNA及蛋白的表达,应用划痕实验和Transwell侵袭实验检测细胞迁移及侵袭能力。结果:siRNA-FOXC1组细胞FOXC1 mRNA和蛋白的表达水平均低于siRNA-NC组和空白对照组(P<0.05)。siRNA-FOXC1组细胞中E-cadherin mRNA及蛋白表达均高于siRNA-NC组和空白对照组,而Vimentin mRNA和蛋白表达均低于siRNA-NC组和空白对照组(P<0.05)。siRNA-FOXC1组迁移率为(13.80±1.25)%,侵袭细胞数为(62.67±5.64)个/高倍视野;空白对照组分别为(40.67±2.10)%和(98.83±4.26)个/高倍视野,siRNA-NC组分别为(42.79±2.48)%和(104.50±7.26)个/高倍视野;siRNA-FOXC1组迁移率和侵袭细胞数均低于空白对照组和siRNA-NC组(P均<0.05)。结论:下调FOXC1表达可能通过抑制上皮间质转化,从而抑制A549细胞的迁移、侵袭能力。 Aim:To explore the changes of migration and invasion ability of lung adenocarcinoma A549 cells after silencing the expression of forkhead box C1(FOXC1).Methods:A549 cells were allocated into blank control group,siRNA-FOXC1 group(transfected with siRNA-FOXC1)and siRNA-NC group(transfected with negative control siRNA).After 6 hours,the mRNA and protein expressions of FOXC1,E-cadherin and Vimentin in the cells were detected by RT-qPCR and Western blot,respectively.And the abilities of cell migration and invasion after 48 hours were measured by scratch and Transwell invasion experiments.Results:The expressions of FOXC1 mRNA and protein in siRNA-FOXC1 group were lower than those in siRNA-NC group and blank control group(P<0.05).Compared with those in siRNA-NC group and the blank control group,the expressions of E-cadherin mRNA and protein in siRNA-FOXC1 group were higher,and those of Vimentin were lower(P<0.05).The migration rate and the number of invasion cells were(40.67±2.10)%and(98.83±4.26)cells/HP in the blank control group,(42.79±2.48)%and(104.50±7.26)cells/HP in siRNA-NC group,(13.80±1.25)%and(62.67±5.64)cells/HP in siRNA-FOXC1 group,and the migration rate and the number of invasion cells in the siRNA-FOXC1 group were lower than those in siRNA-NC group and blank control group(all P<0.05).Conclusion:Down-regulating the expression of FOXC1 may suppress the migration and invasion ability of lung adenocarcinoma A549 cells through inhibiting the epithelial-mesenchymal transition.
