柑橘大实蝇滞育蛹海藻糖酶和山梨醇脱氢酶活性的动态变化 被引量:2

Activity of trehalase and sorbitol dehydrogenase of the Chinese citrus fruit fly,Bactrocera minax(Enderlein)during diapause period

摘要 柑橘大实蝇Bactrocera minax(Enderlein)是我国西南和华中地区柑橘的主要害虫,以滞育蛹在土壤中越冬。为探讨温度对柑橘大实蝇滞育蛹生理状态的影响,本文测定了12、16、20℃和24℃下柑橘大实蝇滞育蛹海藻糖酶和山梨醇脱氢酶活性的变化。结果显示,低温(12℃)下,海藻糖酶活性逐渐上升,高温(24℃)下,海藻糖酶活性逐渐下降;不同温度下山梨醇脱氢酶活性变化不尽相同,在起始阶段(10 d时),柑橘大实蝇滞育蛹山梨醇脱氢酶活性普遍较强,随滞育时间的延长,酶活性出现不同的变化,12℃和20℃下,山梨醇脱氢酶活性呈现“U”形变化趋势,16℃下酶活性逐渐减弱,24℃下酶活性先升后降。16℃下,两种酶活性基本低于同时期其他温度处理。交互作用分析显示,温度和化蛹后天数的交互作用对滞育蛹海藻糖酶和山梨醇脱氢酶活性存在显著影响。上述结果表明,柑橘大实蝇自身生理状态应对不同越冬温度的反应不同,说明越冬温度与柑橘大实蝇滞育期生理水平有着密切的关系。本研究结果对深入了解温度调控柑橘大实蝇滞育期代谢生理机制有一定的参考价值。 Bactrocera minax(Enderlein)is an important citrus pest in southwestern and central China,which overwinters as pupae in diapause in the soil.To investigate the effects of overwintering temperature on the physiological state of the Chinese citrus fruit fly during diapause,the changes in the activity of trehalase and sorbitol dehydrogenase of the Chinese citrus fruit fly overwintering at 12,16,20 and 24℃during diapause period were determined.The results showed that the activity of trehalase gradually increased at lower temperature(12℃),and decreased gradually at higher temperature(24℃).Under different overwintering temperatures,the activity of sorbitol dehydrogenase was different.At the initial stage of diapause(10 d),the activity of sorbitol dehydrogenase was generally higher.With the prolongation of days after pupation,the enzyme activity exhibited different changes.At 12℃and 20℃,the activity of sorbitol dehydrogenase showed a“U”type change trend,and gradually decreased at 16℃,and rose and then dropped at 24℃.At 16℃,the activities of these two enzymes were basically lower than at other temperatures in the same time.Interactive analysis showed that temperatures and days after pupation had a significant effect on the activity of trehalase and sorbitol dehydrogenase of B.minax pupae in diapause.These results indicated that the physiological state of the Chinese citrus fruit fly had different responses to different overwintering temperatures,and the overwintering temperature was closely related to the physiological level of the Chinese citrus fruit fly during diapause period.The present work provides a reference for understanding the regulation of physiological metabolism of the Chinese citrus fruit fly in diapause by overwintering temperatures.
