高通量测序研究中药复方对流感病毒感染小鼠肠道菌群的影响 被引量:10

High-throughput Sequencing Study on the Effects of TCM Compounds on Intestinal Flora of Mice Infected with Influenza

摘要 目的观察麻杏石甘汤、四君子汤、麻杏石甘汤合四君子汤对流感病毒感染小鼠肠道菌群结构和多样性的影响。方法将25只Balb/C小鼠随机分为正常组、病毒组、四君子汤组、麻杏石甘汤组、麻杏石甘汤合四君子汤组,每组5只。建立小鼠亚致死量流感病毒感染模型,除正常组滴鼻生理盐水外,其余各组小鼠用0.1血凝素蛋白(HA)亚致死量病毒稀释液25μL滴鼻感染流感病毒。感染1 h后,正常组和病毒组给予无菌双蒸水灌胃,其余3组给予相应药液灌胃。灌胃第4、7日收集小鼠粪便,采用Illumina Miseq高通量测序检测。结果第4、7日各药物组小鼠肠道菌群Shannon指数较流感病毒感染小鼠变动明显,多样性减少;麻杏石甘汤合四君子汤组病原菌Tyzzerella和颤螺旋菌属明显下降。结论四君子汤、麻杏石甘汤、麻杏石甘汤合四君子汤能改善流感引起的小鼠肠道菌群失调的状态。 Objective To discuss the effects of Maxing Shigan Decoction,Sijunzi Decoction,Maxing Shigan Decoction combined with Sijunzi Decoction on the structure and diversity of intestinal flora in influenza virus-affected mice.Methods 25 Balb/C mice were randomly divided into normal group,virus group,Sijunzi Decoction group,Maxing Shigan Decoction group,Maxing Shigan Decoction combined with Sijunzi Decoction group,with 5 mice in each group.The mice were anesthetized with ether to establish a sublethal influenza virus infection model.The virus group and each administration group were infected with 25μL 0.1 HA sublethal influenza virus dilution by nose drops,and the normal group was given the same amount of sterile saline nose drops.One hour after infection,the normal group and the virus group were given sterile double steaming water for gastric lavage,and the other three groups were given corresponding decoction for gastric lavage.The feces of mice in each group were collected aseptically on the 4th and 7th day of gastric administration.The detection was then carried out with high-throughput sequencing of Illumina Misq.Results On the 4th and 7th days,the Shannon index of the intestinal flora of the mice in each decoction group changed significantly compared with the influenza-infected mice,and the diversity decreased;the pathogenic bacteria Tyzzerella and Treponema in the Maxing Shigan Decoction combined with Sijunzi Decoction group significantly decreased.Conclusion The use of Sijunzi Decoction,Maxing Shigan Decoction,Maxing Shigan Decoction combined with Sijunzi Decoction can improve the imbalance of intestinal flora caused by influenza in mice.
