摘要
为系统挖掘猪沉默信息调节因子5(silence information regulator 5,SIRT5)基因外显子区域SNPs位点,以4个猪资源群体(从江香猪、黔北黑猪♀×巴克夏♂F1代杂交猪、夜郎黄金猪和大约克猪)为研究对象,构建品种DNA池,采用直接测序技术结合生物信息学软件分析猪SIRT5外显子多态性。结果显示:4个猪资源群体中共筛选得到G515A、C831T和A882G 3个变异位点,其中G515A属于错义突变,可由甘氨酸改变为谷氨酸;等位基因频率估算发现,C831T和A882G变异位点在从江香猪群体中多态丰度高,而在大约克猪中均呈现偏态分布;生物信息学发现,G515A突变后,猪SIRT5基因mRNA最小自由能降低,稳定性提升。本研究成功筛选出猪SIRT5基因外显子区的多态位点,为进一步验证该基因突变位点是否具有调控地方猪脂肪代谢水平提供参考。
In order to analyze the polymorphism of the exons of the SIRT5 gene in pigs,four pig resource groups(the Congjiang pig,the Qiangbei black pig×Buckshire pig,the Yelang gold pig,and the Yorkshire pig)were used as the research subjects,and the direct sequencing technology combined with a bioinformatics software was used to mine the SNPs of the exon region of the SIRT5 gene systematically.The results showed that a total of three polymorphic loci(G515 A,C831 T and A882 G)were screened in the four pig resource groups;G515 A belonged to the missense mutation and changed from glycine into glutamic acid at this site.Estimation if the allele frequency showed that the polymorphic abundance of the C831 T an d A882 G mutation sites in Congjiang pig was high,and a skewed distribution was observed in the Yorkshire pig.Bioinformatics analysis showed that,after the mutation of the G515 A site,the minimum free energy of the mRNA of the SIRT5 gene decreased but its stability improved.In this study,the SNPs of the exon region of the SIRT5 gene in pigs was successfully screened,which provided a reference for determination of the function of the gene mutation site in regulating the fat metabolism level of local pigs.
