Effect of Bcl-2-associated athanogene 3 on the proliferation of esophageal cancer cells and its correlation with micro ribonucleic acid let-7

摘要 目的探讨Bcl2相关抗凋亡基因3(BAG3)对食管癌细胞的增殖作用及其与微小核糖核酸let-7(miRNA let-7)的相关性。方法抽取2016年5月至2019年3月焦作市第二人民医院收治的食管癌患者180例,通过手术取患者的食管癌病灶组织及食管正常组织,分别作为观察组与对照组。采用实时荧光定量聚合酶链反应(PCR)技术检测观察组与对照组中BAG3及miRNA let-7表达量。采用person相关系数分析法分析BAG3与miRNA let-7的相关性。通过对食管癌细胞EC109及TE10细胞进行传代培养并转染BAG3,构建过表达及敲低BAG3的EC109及TE10细胞,分别作为BAG3过表达组与敲低BAG3组,监测两组细胞增殖情况。结果观察组BAG3表达量高于对照组(P<0.05)。观察组miRNA let-7低于对照组(P<0.05)。BAG3蛋白与miRNA let-7呈负相关。BAG3过表达组增殖速度高于敲低BAG3组。结论BAG3在食管癌病灶组织中高表达,在食管正常组织中低表达,具有促进食管癌细胞增殖作用。食管癌中BAG3与miRNA let-7呈负相关,敲低BAG3表达量可抑制肿瘤细胞增殖。 Objective To investigate the effect of Bcl-2-associated athanogene 3 (BAG3) on proliferation of esophageal cancer cells and its correlation with micro ribonucleic acid let-7(miRNA let-7).Methods In this study, the cancer tissues (observation group) and adjacent normal tissues (control group) were collected from 180 patients with esophageal cancer treated in the Second People’s Hospital of Jiaozuo from May 2016 to March 2019. The expressions of BAG3 and miRNA let-7 in the observation group and the control group were detected by real-time fluorescence quantitative polymerase chain reactio(PCR). The correlation between BAG3 and miRNA let-7 was determined by Pearson correlation analysis. EC109 and TE10 cells were cultured and transfected with overexpressed BAG3 and knocked down BAG3, as BAG3 overexpression group and the BAG3 knockdown group. Then the cell proliferation of the two groups was detected.Results The expression of BAG3 in the observation group was significantly higher than that in the control group (P<0.05). The miRNA let-7 in the observation group was significantly lower than that in the control group (P<0.05). BAG3 protein was negatively correlated with miRNA let-7. The proliferation rate of BAG3 overexpression group was higher than that of BAG3 knockdown group.ConclusionsBAG3 is highly expressed in the cancer tissues and lowly expressed in adjacent normal tissues, indicating its promotion effect on esophageal cancer cells. Meanwhile, the BAG3 protein is negatively correlated with miRNA let-7, so knocking down the expression of BAG3 can inhibit the proliferation of cancer cells.
