FOXC1和lncRNA FOXCUT在结直肠癌患者中的表达及其对病情进展的影响 被引量:10

Expression of FOXC1 and lncRNA FOXCUT in colorectal cancer patients and its effect on disease progression

摘要 目的 探讨叉头框基因(FOXC1)和长链非编码RNA(lncRNA)启动子临近转录体(FOXCUT)在结直肠癌(CRC)患者中的表达及其对病情进展的影响.方法 选取2013年1月至2014年1月在郑州大学附属郑州中心医院行CRC根治切除手术的65例患者的癌组织和癌旁组织标本,采用实时聚合酶链式反应法测定FOXC1和lncRNA FOXCUT在其组织中的表达情况,并分析FOXC1和lncRNA FOXCUT表达情况对病情的影响.结果 癌组织中FOXC1和FOXCUT表达均高于癌旁组织(P<0.05);患者CRC组织中FOXC1和FOXCUT表达与性别、年龄、肿瘤部位无明显关系(P>0.05),与肿瘤直径、TNM分期、肿瘤浸润深度、远处转移情况有明显关系(P<0.05).肿瘤直径、TNM分期、肿瘤浸润深度、远处转移、FOXC1表达、FOXCUT表达是结直肠癌患者预后的独立影响因素(P<0.05).结论 FOXC1和FOXCUT与CRC肿瘤分期、肿瘤直径、转移情况、浸润深度等有密切关系,可能参与CRC的发生与发展,可作为CRC患者预后的分子标志物. Objective To investigate the expression of forkhead box C1(FOXC1)and long non-coding RNA(lncRNA)FOXC1 promoter upstream transcript(FOXCUT)in the colorectal cancer(CRC)patients and its effects on the disease progression.Methods In this study,the CRC tissues and adjacent tissues were collected from 65 CRC patients in Zhengzhou Central Hospital Affiliated to Zhengzhou University from January 2013 to January 2014.The expression levels of FOXC1 and lncRNA FOXCUT were detected by real-time polymerase chain reaction,and their effects on the disease progression were analyzed.Results The expression levels of FOXC1 and lncRNA FOXCUT of the cancer tissues were higher than those in the adjacent tissues(P<0.05).The expression levels of FOXC1 and lncRNA FOXCUT in the CRC tissues were not related to the gender,age or tumor location(P>0.05),while they were significantly correlated with the tumor diameter,tumor nodes and metastasis(TNM)staging,invasion degree and distant metastasis(P<0.05).Tumor diameter,TNM staging,invasion degree,distant metastasis,FOXC1 expression,and FOXCUT expression were the independent prognostic factors for CRC patients(P<0.05).Conclusions Expression levels of FOXC1 and FOXCUT are closely related to the tumor staging,tumor diameter,metastasis and invasion degree of CRC patients.FOXC1 and FOXCUT may be involved into the occurrence and development of CRC,and they can be used as prognostic factors for CRC patients.
