响应面法优化浸提芦苇叶黄酮工艺条件的研究 被引量:1

Study of Optimizing Extraction Conditions of Flavonoids from Phragmites Leaves by Response Surface Methodology

摘要 通过响应面法对乙醇水溶液浸提芦苇叶黄酮的工艺条件进行优化,以提高黄酮的产率;通过扫描电镜探究浸提法对芦苇叶结构的影响,并从植物结构的角度对提取过程机理进行探究。以料液比、加热温度、加热时间、乙醇体积分数作为影响黄酮提取的单因素进行试验,建立四因素三水平的响应面数据模型,获得最佳工艺条件为:加热温度为59.9℃,加热时间为1.995 h,料液比为1:25.13(g/mL),乙醇体积分数为59.7%,此时,黄酮的收率为24.310 mg/g;由扫描电镜图分析得到浸提法对芦苇的结构具有很强的影响作用,通过改变芦苇叶片的结构将其中的物质提取出来。 The optimum technological conditions of flavonoids extracted from ethanol aqueous solution were optimized by using the response surface method in order to achieve the maximum yield of flavonoids.The effect of leaching formulation on the structure of Reed leaves was explored by using scanning electron microscope,and the mechanism of extraction process was investigated from the point of view of plant structure.Based on the single factor test of the material liquid ratio,heating temperature,heating time and ethanol concentration,the four-factor three-level response surface data model was established.The optimum technological conditions were heating temperature 59.9℃,heating time 1.995 h,material liquid ratio 1∶25.13 g/mL,and ethanol concentration 59.7 per cent,with thhe yield of flavonoids being 24.310 mg/g.By scanning electron microscopy,the leaching formula had a strong effect on the structure of reed,and the material was extracted by changing the structure of reed leaves.
