摘要
目的以丹参素含量为指标,优化中药复方的提取工艺条件.方法建立HPLC法测定丹参素的含量方法;对浸泡时间进行单因素考察,确定其最佳浸泡时间和药材吸水率;采用L9(34)正交实验设计,以丹参素的总含量和浸膏得率为指标,考察加水量、煎煮次数和煎煮时间3个因素对提取结果的影响.结果HPLC法测定丹参素的含量的最佳条件为:色谱柱:Inertsil ODS-3(4.6 mm×250 mm,5μm);流动相:甲醇-0.01%冰醋酸水溶液(7:93);流速:1 mL/min;检测波长:281 nm.丹参素的最佳提取工艺为:加水煎煮3次,加8倍量水,每次煎煮1h.结论按正交设计试验法优化的工艺条件提取"参经颗粒"中药复方,工艺可行,浸膏质量稳定.
Objective to optimize the extraction process of traditional Chinese medicine compound with danshensu content as the index.Methods HPLC method was established for the determination of danshensu.The best soaking time and water absorption of medicinal materials were determined by single factor investigation.L9(34)orthogonal experimental design was used to investigate the influence of three factors,namely,the amount of water added,the number of frying and the time of frying,on the extraction results.Results the best condition for determination of danshensu content by HPLC was Inertsil ODS-3(4.6 mm×250 mm,5 microns).Mobile phase:methanol-0.01%glacial acetic acid solution(7:93);Flow rate:1mL/min;Detection wavelength:281nm.The best extraction technology of danshensu is:add water and cook for 3 times,add 8 times of water,and cook for 1h each time.Conclusion it is feasible to extract shenjing granules from traditional Chinese medicine compound by orthogonal design.
