摘要
目的观察靶向多肽SS-31对载脂蛋白E(ApoE)基因敲除动脉粥样硬化大鼠的干预效果。方法2018年12月—2019年4月于中国人民解放军空军军医大学第二附属医院实验动物中心进行实验。ApoE基因敲除雄性大鼠60只,按照随机数字表法分为2组,各30只。实验组大鼠皮下注射SS-31,对照组大鼠皮下注射同等剂量的生理盐水。2组大鼠均高脂饲养10周后处死,取主动脉窦染色,比较2组大鼠主动脉斑块面积及大小,取血液样本检测血清三酰甘油(TG)、总胆固醇(TC)、血清细胞内黏附因子-1(ICAM-1)、单核细胞趋化因子-1(MCP-1)水平,观测大鼠主动脉切片活性氧(ROS)、超氧化物歧化酶(SOD)及ATP水平,免疫组化法检测大鼠CD36及LOX-1的表达水平。结果实验组大鼠主动脉斑块面积及大小均低于对照组(t/P=5.664/<0.001,5.035/<0.001);血清ICAM-1及MCP-1水平均低于对照组(t/P=8.138/<0.001、5.253/<0.001),而2组TG、TC水平比较差异无统计学意义(P>0.05)。实验组主动脉组织ROS及SOD水平明显低于对照组,ATP水平明显高于对照组(t/P=7.533/<0.001、2.580/0.018、8.794/<0.001);CD36及LOX-1表达均显著低于对照组(t/P=5.957/<0.001、7.170/<0.001)。结论靶向多肽SS-31对ApoE敲除大鼠动脉粥样硬化具有较好的治疗效果,与该药能够缓解大鼠氧化应激反应有关。
Objective To observe the intervention effect of targeting peptide SS-31 on ApoE gene knockout atherosclerosis rats.Methods From December 2018 to April 2019,the experiment was conducted in the experimental animal center of the Second Affiliated Hospital of the Air Force Medical University of the People's Liberation Army.Sixty male ApoE knockout rats were divided into two groups,30 rats in each group.SS 31 was injected subcutaneously in the experimental group and saline of the same dose was injected subcutaneously in the control group.The rats in both groups were sacrificed after 10 weeks of high-fat feeding.The area and size of aortic plaque were compared.Blood samples were taken to detect serum TG,TC,ICAM-1 and MCP-1)The levels of ROS,SOD and ATP were measured.The expressions of CD36 and LOX-1 were detected by immunohistochemistry.Results The area and size of aortic plaque in the experimental group were lower than those in the control group(t/P=5.664/<0.001,5.035/<0.001);the levels of serum ICAM-1 and MCP-1 were lower than those in the control group(t/P=8.138/<0.001,5.253/<0.001),but there was no significant difference in TG and TC levels between the two groups(P>0.05).The level of ROS and SOD in the experimental group was significantly lower than that in the control group,and the level of ATP was significantly higher than that in the control group(t/P=7.533/<0.001,2.580/0.018,8.794/<0.001);the expression of CD36 and LOX-1 was significantly lower than that in the control group(t/P=5.957/<0.001,7.170/<0.001).Conclusion Target peptide SS 31 has a good therapeutic effect on atherosclerosis in ApoE knockout rats,which is related to its ability to alleviate oxidative stress in rats.
